Dat klopt wel met het koersverloop van Galapagos.
Dan komt ook gelijk de vraag:
Heeft het bestuur van Galapagos met opzet een (te) positieve outlook gegeven om de koers op te jagen met als doel een extra aandelenuitgifte?
Natuurlijk hebben ze een positieve outlook gegeven en natuurlijk kunnen positieve outlooks nuttig zijn voor de financiering. Moeten blij wezen dat ze wat dat betreft niet timide zijn, want anders stond het bedrijf volgens mij niet waar het staat. Met opzet een "te" positieve outlook is weer een ander verhaal. Dan gaat richting een zware beschuldiging en zo'n beschuldiging moet niet geuit worden zonder bewijs. Ook moet niet worden vergeten dat niet elke vertraging van elk onderzoek valt onder informatie die meteen moet worden gedeeld tijdens een halt of iets dergelijks.

De Therapeutics Development Network die bij de CFF hoort heeft zoals je weet een grote invloed op de participatie en prioritering van onderzoeken.
Meerdere onafhankelijke experts bekijken de data van in dit geval de verschillende CFTR modulatoren. Aan de hand van verschillende criteria maken ze een algemeen plaatje. Er wordt een score toegekend wat de TDN sites moet helpen bij het maken van hun keuzes.
CFF doet zaken met verschillende bedrijven en is er echt wel van bewust dat ze moeten waken voor onethische praktijken. Hoe dat in casu het geval zou kunnen zijn? Zoals jij zie ik het niet.
Asti, volgens mij denken we er hetzelfde over. Dat is ook precies wat ik probeerde te schrijven:Overigens zijn de suggesties dat CFF niet onafhankelijk zou zijn en mogelijk zelfs eigen gewin boven patiënten zou stellen zware beschuldigingen. Graag zou ik wat extra motivatie daarvoor zien. Ik nodig juist uit tot het beargumenteren van de verwijten/beschuldigingen aan het adres van CFF. De analisten hebben zich hier met een kluitje het riet in laten sturen door de CSO.

Ik ben niet ingegaan op de inconsistenties in de communicatie over de voortgang van het CF-programma. Anderen, waaronder met name aston.martin, hebben dat gedaan.
Ik ben het er overigens volledig mee eens dat transparantie vanuit Galapagos een groot goed is. Maar soms ook is spreken over bepaalde zaken ‘zilver’, en zwijgen ‘goud’. Wanneer je
vertrouwen hebt in het management, accepteer je die (tijdelijke) keuze. Ik doe dat in ieder geval, totdat het tegendeel blijkt. Van de Stolpe en zijn collega’s kunnen vooralsnog bogen op een uitstekend ‘track record’.

Hoewel ik niet heb berekend hoeveel Onno aan vremogensrendementsbelating moet betalen over zijn bezit aan aandelen en warrants in Galapagos, maar zijn afrekening over 2016 zal al niet mals zijn en over 2017 kan hij nu ook beter dit jaar gaan betalen ter voorkoming van heffingsrente. Ik zou er maar niets achter zoeken achter die verkoop van 15,000 aandelen op 20 juni. Hij heeft alleen het minimaal verplichte deel van zijn warrants uitgeoefend terwijl hij er ook meer had kunnen uitoefenen.

Maar dit wantrouwen is wel wat je krijgt als je (de onderneming) niet transparant bent. Vertrouwen komt te voet en gaat te paard
Wat mij betreft krijgt management nog steeds voordeel van de twijfel. Gezien betrokkenheid bij dit forum/beleggers in het verleden kan ik me voorstellen dat men graag volledige openheid van zaken zou willen geven echter de zaken( o.a. wijze van communiceren) zijn veranderd sinds de partnering met Gilead en ook de voortgang met AbbVie.

Als je veel geld hebt zitten in aandelen die geen dividend opleveren maar wel in waarde stijgen is het moment van fiscale afrekening cashflow matig een vervelend moment. Vermogen tijdig overhevelen naar de volgende generatie levert ook niet veel soelaas op. Gelukkig is voor mij de resulterende meer bescheiden aanslag nog wel de komende week te betalen uit mijn cashflow.

Dank @ charel 1 Op 26 juni 2017 hebben de analisten van Goldman Sachs hun beleggingsadvies voor Galapagos (GLPG; ISIN: BE0003818359) herhaald. Het advies van Goldman Sachs voor Galapagos blijft "kopen". Het koersdoel wordt door Goldman Sachs verlaagd van 96,00 EUR naar 94,00 EUR.
Goldman Sachs June 23 2017Updating numbers for cystic fibrosis delay What's changed At its recent R&D day, Galapagos highlighted its intention to pursue at least three proprietary triple combinations for the treatment of Cystic Fibrosis (CF). We view the additional shots on goal as a positive. However, as a consequence of planned discussions with UK regulatory authorities around best trial design, its previous guidance of a mid-2017 triple combination trial initiation has now been revised to 4Q17. Because of this delay and the plan to initiate multiple triple combination studies, we revise our forecasts to model the launch of CF in late 2021 (previously 2020). AbbVie has the rights to Galapagos’ cystic fibrosis compounds, and Galapagos receives royalties in the mid-teens to 20% and milestones. Implications As a result of this slight delay in timeline, the value of the CF indication in our DCF-based SOTP valuation decreases to €7/share (previously €9/share), resulting in a new 12-month price target of €94 (from €96).
Janney June 21 2017Flash Takeaways Based on the most recent CF-portfolio guidance of an anticipate triple-combo clinical program start during the summer, investors headed into the R&D event expecting clarity on trial design and affirmation on timelines. Unfortunately, the triple-combo clinical program start has now been pushed into 4Q awaiting clear regulatory guidance. On the positive side, the CF portfolio has multiple shots on goal and when the program does get the green light, Galapagos is planning to execute on multiple triple-combinations, mimicking competitor Vertex (VRTX - Neutral). Recall, VRTX is expected to roll out triple-combo data starting 3Q17, the delays put Galapagos more than a year behind. Furthermore, based on the multiple combinations being evaluated, Galapagos and partner AbbVie (ABBV – No rating) may not be able to finalize candidate selection for the pivotal program until 2019, by which time we suspect Vertex could be half-way through with theirs. On a more optimistic note, GLPG is expected to report data from the ALBATROSS (4Q17) and FLAMINGO (1Q18). Since both ALBATROSS (corrector '2222 + ivacaftor) and FLAMINGO (corrector '2222 monotherapy in F508del homozygous patients) we suspect that neither of these are likely to be stock moving. Other near-term data milestones include: preliminary data from its wholly-owned GLPG-1690 in IPF patients during 3Q17 and topline data from MOR106 (atopic dermatitis) during 4Q17. Note, Gilead (GILD- No rating) partnered filgotinib continues to hum along with multiple registrational and phase 2 studies with data rollout begining mid-2018, by our estimates. However, the blockbuster potential of filgotinib is adequately reflected in the stock, in our view, and we await better entry points to get more constructive on an otherwise compelling pipeline.

Op dit inhoudelijke forum hebben verschillende mensen, waaronder aston.martin en Reykus, hun mening gegeven over de ontwikkelingen van de laatste week, waarvoor veel dank. De consensus lijkt (ruwweg) te zijn: er is een tegenvaller in het CF tijdspad en de communicatie daarover verdient niet de schoonheidsprijs, maar fundamenteel blijft alles er voor Gala gewoon prima uit zien. Ook de reacties van analisten lijken gematigd. Wat ik echter nog gemist heb is een reactie van een van onze eigen grote analisten, pe26. Zou jij wellicht nog even kort kunnen aangeven hoe je nu, een week na data, aan kijkt tegen de stand van zaken?

Voor wat het waard is, de vertaling door Goldman Sachs van de CF vertraging/tegenvaller in Euro's: De waardering van CF in de Galapagos koers gaat van €9/share naar €7/share. Dat is een waardedaling van zo'n 20% in het CF programma. Op de totale waarde van Galapagos, die voor het overgrote deel door filgotinib wordt bepaald is het €2/share, oftewel zo'n €100 000 000, oftewel zo'n 3%. De 'sentiment' schade in Euro's lijkt me groter. Was Galapagos tot voor kort een hemelbestormend aandeel waarin niets fout leek te kunnen gaan en de bomen tot in de hemel rezen, nu blijkt weer eens dat de koers ook flink omlaag kan, en dat er in de pipeline van Galapagos niet alles vanzelf goed gaat. Het kan weer even gaan duren voordat het sentiment 1) bijtrekt naar normale nivo's, en 2) doorstijgt naar het nivo gouden bergen. Ik verwacht nivo 1) nog dit jaar, mede afhankelijk van start 1e triple combo, en nivo 2) later in 2018 door eerste rapportages filgotinib phase III plus overnamekoorts in verband met aflopen standstill.

Klopt, niet precies bekend. Maar wel ongeveer. Veel mensen hier, waaronder ik, gaan uit van 3 jaar. Mede ook vanwege het interview (zie onderaan). De deal met Gilead is ingezet eind 2015, en de aandelen/deal en de extra bak Gilead geld is ge-effectueerd in begin 2016. Dus ik schat in dat de standstill eind 2018 / begin 2019 afloopt. Het kan ook 2 1/2 jaar zijn. 4 jaar lijkt me teveel. Eerder korter dan langer dan 3 jaar. Ik denk dat velen met mij (de markt ) aan deze periode denken. En dat de overnamekoorts niet van het ene op het andere moment opkomt, maar langzaam gaat oplopen in 2018, met lichte verhoging aan het begin van de zomer, en roodgloeiend richting jaareinde 2018. We have previously demonstrated a crucial role for ß-catenin signaling in regulating LSCs and identified GPR84 as an important ß-catenin regulator in the maintenance of mixed-lineage leukemia (MLL) LSCs (Wang et al., Science 2010; Dietrich et al., Blood 2014). Hence, targeting LSCs by pharmacological inhibition of GPR84/ß-catenin signaling represents a promising therapeutic approach. In collaboration with a pharmaceutical company that has developed a novel GPR84 antagonist (GP), we investigated the effect of GP in MLL pre-leukemic stem cell (pre-LSC) function. GP (20 µM) significantly inhibited the colony forming ability of MLL pre-LSCs (P < 0.0001) but had little effect on normal hematopoietic stem cells. Quantitative RT-PCR and western blot analysis confirmed GP-induced downregulation of GPR84 target genes, including Hoxa5, Hoxa7 and Meis1a, indicating GP-induced inhibition of GPR84 signaling. To further examine the mechanism of GPR84 inhibition on MLL pre-LSCs, we evaluated several epigenetic regulators (i.e. JMJD1c and EZH2) known to promote leukemogenesis (Zhu et al., ?J Clin Invest 2016; Tanaka et al., Blood 2012). Western blot analysis showed that inhibition of GPR84 signaling did not alter the expression of JMJD1c or EZH2. However, we observed a significant increase in the expression of a novel and not-yet-characterized histone demethylase (HD) in AML. To investigate the role of HD in AML leukemogenesis, we overexpressed HD in MLL pre-LSCs and subsequent serial replating assay showed a marked reduction in colony forming ability (P < 0.005), indicating impaired self-renewal in vitro. Consistent with our in vitro observations, in vivo transplantation in syngeneic mice revealed a significant delay in leukemia onset and increase in mouse survival (P < 0.001). We next performed western blot analysis to examine the demethylase activity of HD, and our data revealed that HD overexpression caused a substantial reduction in global histone 3 lysine 36 dimethylation (H3K36me2), an epigenetic mark normally associated with transcriptional activation and elongation. In order to identify genes regulated by HD through demethylation of H3K36me2, we performed H3K36me2 ChIP-seq on HD overexpressing MLL pre-LSCs. Our analysis identified several genes including anti-apoptotic protein Mcl-1 and angiogenic receptor Nrp1, which are known to be involved in AML leukemogenesis, with decreased H3K36me2 mark on both the transcriptional start site and gene body. Subsequent western blot analysis confirmed the decreased expression of both Mcl-1 and Nrp1 in HD overexpressing pre-LSCs. Given the prominent roles of anti-apoptosis and angiogenesis in the development of hematologic malignancies such as leukemia, we are currently evaluating these mechanisms caused by HD overexpression in an important subtype of AML.
