    argenx announces publication of seminal data supporting the therapeutic potential of ARGX-110 for acute myeloid leukemia in the Journal of Experimental Medicine



    Breda, the Netherlands / Ghent, Belgium - argenx (Euronext Brussels: ARGX), a clinical-stage biopharmaceutical company focused on creating and developing differentiated therapeutic antibodies for the treatment of cancer and severe autoimmune diseases, today announced the publication of new preclinical data on the CD70/CD27 pathway that provide further rationale for ARGX-110 therapy for the treatment of acute myeloid leukemia (AML). ARGX-110, a SIMPLE Antibody(TM) targeting CD70, is currently being evaluated in a Phase I/II study in combination with azacitidine in newly diagnosed AML patients.



    Data published by argenx collaborator Prof. Adrian Ochsenbein at the University of Bern, demonstrate that CD70/CD27 is highly expressed on AML blasts and leukemic stem cells in newly diagnosed AML patients, regardless of cytogenetic factors, or the patient's risk class. The published data show the CD70/CD27 pathway to be critical in the biology of leukemic stem cells, and therapeutic intervention using a CD70-targeted antibody to enable selective targeting of leukemic stem cells without impacting hematopoietic stem cells, resulting in a survival benefit in preclinical AML models.

    About ARGX-110

    ARGX-110 is a SIMPLE Antibody(TM) targeting CD70, an immune checkpoint target involved in hematological malignancies, several solid tumors and severe autoimmune diseases. ARGX-110 works in three ways: i) blocks growth of tumor cells, ii) kills cancer cells and iii) restores immune surveillance against tumors (Silence K. et al. mAbs 2014; 6 (2):523-532). ARGX-110 is currently being evaluated in hematological and solid tumors. Preclinical work on ARGX-110 in AML was done in collaboration with the Tumor Immunology Lab of Prof. A. F. Ochsenbein at the University of Bern. Prof. Ochsenbein won the prestigious 2016 Otto Naegeli Prize for their breakthrough research on CD70/CD27 signaling with therapeutic potential for cancer patients.



    About AML

    AML is a neoplasia of the myeloid line of blood cells, characterized by the rapid growth of abnormal white blood cells that build up in the bone marrow and interfere with the production of normal blood cells. AML has the lowest 5-year survival rate of all blood malignancies (~5-20%) and primarily affects the eldery. Patients above 65 years old are unfit for stem cell transplantation and are often put on palliative treatment. Around 40% of these patients receive azacitidine, as it has been shown to improve the overall survival rate.

    Gefeliciteerd Fly .Inderdaad . Winst is winst . Dat dacht ik echter ook bij GLPG toen ze op 17 stonden .1,5 jaar later 60+

    Bij mij blijft het stevig op de plank . Ik verwacht dit jaar een (zeer )grote deal .
    Ik niet alleen , want de koers loopt langzaam maar gestaag op . Totale beurswaarde is nog zeer laag terwijl er een paar blockbuster tegen de horizon aan gluren .
    Celgene zou zo maar een zeer goede kandidaat kunnen zijn .

    Heeft iemand toevallig een company presentation met gegevens voor 2017? De laatste versie op de site is van november, waarin 2017 ontbreekt.

    ARGX-113 resultaten van Ph1 heb ik nog niet voorbij zien komen, Ph2 moet ook nog starten.

    Zijn er eigenlijk banken/investeringsmaatschappijen die argenx volgen?

    Piper Jaf­fray. Die plakt een koop­ad­vies en een koers­doel van 20 euro op het aan­deel. Geen en­ke­le an­de­re ana­list heeft zo'n hoog koers­doel.

    Het gaat het bi­o­tech­be­drijf voor de wind. Het be­drijf sloot eer­der dit jaar een grote al­li­an­tie met far­ma­reus Abb­Vie voor de ont­wik­ke­ling van nieu­we kan­ker­be­han­de­ling. Het kreeg daar­voor met­een 40 mil­joen dol­lar, en daar kan - als het on­der­zoek vlot ver­loopt- nog 645 mil­joen dol­lar bij­ko­men. De deal was de eer­ste, grote er­ken­ning voor de tech­no­lo­gie van het Gent­se bi­o­tech­be­drijf. Met het goede nieuws op zak, tank­te het be­drijf nog eens 30 mil­joen dol­lar vers ka­pi­taal­bij, voor­na­me­lijk bij en­ke­le grote Ame­ri­kaan­se in­ves­teer­ders. De ana­list van Piper Jaf­fray schat dat Ar­genx nu vol­doen­de cash heeft (zo'n 118 mil­joen euro) om zijn on­der­zoek tot in 2018 te blij­ven fi­nan­cie­ren. Tijd ge­noeg dus om te be­wij­zen dat zijn on­der­zoek waar­de­vol is.

    Het beurs­huis is voor­al en­thou­si­ast over de zoek­tocht van Ar­genx naar een me­di­cijn tegen my­asthe­nia gra­vis, een zeld­za­me spier­ziek­te die le­vens­be­drei­gend kan zijn. Het is een au­to-im­muun­ziek­te, ge­ken­merkt door op­sto­ten van spier­zwak­te. Voor­al veel­ge­bruik­te spie­ren wor­den aan­ge­val­len, zoals tong-, kauw-, oog- en adem­ha­lings­spie­ren. Het on­der­zoek zit nog een in een vroeg sta­di­um. Het mid­del is enkel nog maar ge­test op ge­zon­de proef­per­so­nen.

    argenx launches Phase II proof-of-concept study of ARGX-113 for the treatment of myasthenia gravis

    Breda, the Netherlands / Ghent, Belgium - argenx (Euronext Brussels: ARGX), a clinical-stage biopharmaceutical company focused on creating and developing differentiated therapeutic antibodies for the treatment of cancer and severe autoimmune diseases, today announced the initiation of a Phase II proof-of-concept study of ARGX-113 in patients with myasthenia gravis (MG).



    "MG is a rare and debilitating muscle disease with limited effective and sustainable treatments. ARGX-113 has the potential to eliminate patient symptoms while minimizing common side effects seen with current treatments by reducing the pathogenic IgG levels," commented Nicolas Leupin, CMO argenx. "The initiation of this Phase II study is an important milestone in understanding how ARGX-113 can be effective in a wide range of IgG-mediated autoimmune diseases including additional orphan indications, and larger indications like multiple sclerosis and lupus. We expect to start a Phase II trial in a second orphan IgG driven indication, immune thrombocytopenia (ITP), by the end of the first quarter of 2017."



    The double-blind, placebo controlled Phase II study will enrol up to 24 MG patients with confirmed generalized muscle weakness. ARGX-113 will be dosed on top of current standard of care, corticosteroids and/or immunomodulatory agents. The primary endpoints of the trial are safety and tolerability and secondary endpoints include efficacy, impact on quality of life and an assessment of pharmacokinetics (PK) and pharmacodynamic (PD) markers.



    In Phase I clinical trials, ARGX-113 demonstrated favorable safety and tolerability across multiple doses and dosing regimens with promising pharmacodynamics effects relating to speed, depth and duration of IgG reduction.

    ARGX-113 is a potential breakthrough therapy for treatment of IgG-mediated autoimmune diseases. ARGX-113 is the Fc-portion of an antibody that has been modified by the argenx proprietary ABDEG(TM) technology to increase its affinity for FcRn beyond that of normal IgG antibodies. As a result, ARGX-113 blocks antibody recycling and leads to fast depletion of the autoimmune disease-causing IgG autoantibodies. The development work on ARGX-113 is done in close collaboration with Prof. E.     Vooral onderstaande regel lijkt me een belangrijke . Kan effectief zijn in een groot aantal uiteenlopende ziektes . MS en LUPUS zeer belangrijk .

    Moii persbericht; helaas heeft Tim vorige week weer verkocht. Kan zijn gedrag niet rijmen maar heeft naast zijn riante salaris blijkbaar toch behoefte om extra te cashen en zijn goedkoop verkregen opties snel te verzilveren. Blijf er toch maar even uit maar blijf wel volgen want waardering is nog niet al te hoog en platform blijft zeer interessant.
