De pijplijn van Affimed is inmiddels groter en zit al in fase 2. Enkel de kas bij Fate is een stukje groter. Waarom is Fate nog steeds meer dan 2 keer zo veel waard?

@Ontop1, generaliseren is niet mijn ding. Hoess zijn rijkdom is grotendeels onderbelicht maar ik ben er vrij zeker van dat Scott Wolchko van Fate (naast Stanley C. Erck van Novavax ) inmiddels met stip op het erepodium staat van meest gehate CEO in biotechland in 2022/-23. Beiden hebben zich buitensporig verrijkt door tijdige verkoop van aandelen die ze voor een krats dmv personeelsopties konden verwerven.

Tja van $ 11 miljard marktkapitalisatie terug naar $ 400 mio (wat nog aan de hoge kant is in mijn beleving) is natuurlijk een kleine ramp die voorkomen had kunnen worden als al die goed betaalde analisten beter hadden opgelet. Nu het leed is geschied komen ze op een enkeling na achteraf op het laatste moment met draconische koersdoelaanpassingen. Lijkt me ook een harde les voor die "zieners" van Cathie Wood (met een aandeel van 11%!) en een onbekende grootheid als Redmile. Zelfs vriend Faison zegt op 5 januari 2023 in een artikel op Seeking Alpha dat hij met de aandelen die hij in 2022 terug kocht een verlies van 68% geleden heeft, die had het dus ook niet zien aankomen. Kortom collectieve hebzucht en onvoldoende kennis van zaken. In het recente Pinto/AFMD artikel wordt alleen gezegd dat die onsplijtbare CD16 receptor voorwerp van een wetenschappelijk debat is. Een echte wetenschapper is natuurlijk voorzichtig met zijn uitspraken. Ik denk dat die Engelsen en Zweden het echter goed onderzocht hebben en de radicale omzwaai bij Fate is een teken van hun gelijk.

Affimed expects to report data from all three ongoing AFM24 studies at scientific conferences in Q2/Q3 2023. AFM24-101: Affimed continues to enroll patients in the expansion phase of the AFM24 monotherapy study at the RP2D. The expansion cohorts include patients with renal cell carcinoma (clear cell), non-small cell lung cancer (EGFR mutant) and colorectal cancer. AFM24-102: Enrollment is completed in the 480 mg dose escalation cohort of the phase 1/2a combination study of AFM24 with the anti-PD-L1 checkpoint inhibitor atezolizumab (Tecentriq®) in patients with advanced epidermal growth factor receptor-expressing solid tumors. AFM24-102 includes patients with non-small cell lung cancer (EGFR wildtype), gastric and gastroesophageal junction adenocarcinoma and pancreatic/hepatocellular/biliary tract cancer. No dose-limiting toxicities (DLTs) have been observed thus far in the dose escalation. Expansion cohorts are expected to begin enrollment in Q1 2023. Data from the first cohort (4 patients at 160 mg dose) of the phase 1 dose escalation study presented at the annual meeting of the Society for Immunotherapy of Cancer (SITC) in November 2022 showed that clinical activity was observed in two patients. A patient with gastric cancer and skin metastases who had rapidly progressed following four prior lines of therapy, including a PD-1 inhibitor, achieved a partial response. A second patient with pancreatic adenocarcinoma showed stable disease beyond four months. Patients being enrolled in the study are required to have progressed or relapsed on standard of care therapies. AFM24-103: In the phase 1/2a combination study of AFM24 with SNK01, NKGen Biotech’s ex vivo expanded and activated autologous NK cell therapy, enrollment has been completed in the dose cohort of 480 mg AFM24 weekly, with no DLTs observed to date. Ik vermoed/hoop dat Genentech daarover deze week iets gaat zeggen en dan maai je het gras niet weg voor je grote partner.
Tenminste geen terugtrekkende bewegingen van partners zoals bij Fate. Ook de cash runway blijkt verlengd te zijn met een half jaar zonder ontslagen. Affimed weet haar middelen een stuk efficiënter te besteden dan Fate dat meer dan het dubbele in kas heeft (en even veel personeel). Fate heeft haar (ongetwijfeld) dure allianties met de Universiteit van Minnesota en het Memorial Sloan Kettering Cancer (dat geleid wordt door CAR-T coryfee Michael Sadelain) natuurlijk nog wel op de pay list. Fate zegt gedekt te zijn tot eind 2025, maar die zitten zeker tot die datum nog steeds in fase 1. Het wordt tijd dat de waardering van AFMD minimaal op het zelfde niveau als dat van Fate wordt gebracht.
De longs lijken zich weer vergist te hebben, dacht men dat op 9 januari er door de FDA al een kant en klaar besluit over AFM13 + Artiva zou zijn genomen? En Fate stijgt bijna 14%????

Misschien een concretere uitspraak dan het algemene 'Based on the written response, Affimed remains on track to submit an IND in the first half of 2023 and, subject to FDA clearance of the IND, to initiate a clinical study during 2023.' Waarmee het nog alle kanten (lees: de lange en verkeerde) op kan.
Of men denkt dat er nog kansen genoeg gaan komen. Affimed blinkt niet uit in snelheid van handelen, dan komen op de lange duur nog wel kapitaalverhogingen in zicht met de nodige verwatering. Ik hou mij maar vast aan de resultaten, teksten zoals: "The combination treatment continues to show a 100% overall response rate (ORR) at the highest dose level and a further improvement in the complete response (CR) rate, from the previously reported 62% to 70.8%." Oftewel, komt op den duur wel goed, maar wat achter de hand houden om bij te kopen op nog lagere koersen kan vast geen kwaad.

Ja dat zou wel eens de enige geldige conclusie kunnen zijn. Het past ook bij de uitspraak dat er nu cash is tot 2025. Dat zou dan niet eens voldoende zijn voor een nieuw fase2 onderzoek. Een overname komt steeds dichterbij. Ik denk dat er wel een Europese oplossing zou kunnen komen. De eerst gegadigde is natuurlijk Roche, anders had men net als Janssen bij Fate, de pijp wel aan Maarten gegeven. Maar er zijn meer . Zo heeft Merck KgaA een gevestigde marktpositie met Erbitux, een monoclonaal antilichaam dat zich vastmaakt op EGFR . AFM24 zou hier naadloos bij passen. En Sanofi , die Kiadis voor $358 miljoen heeft over genomen voor haar NK cellen, zou ik ook niet helemaal willen uitsluiten. Sanofi heeft ook net nog een deal gesloten met Innate Pharma gericht op B7H3. Het is in feite een voortzetting van een samenwerking die al in 2016 begon. Sanofi is er dus van overtuigd dat de ANKET technologie (een broertje van de ICE technologie) +NK cellen wel degelijk toekomst hebben. Wellicht streeft Hoess er naar om van Affimed een 2e Innate Pharma te maken maar het duurt allemaal zo verschrikkelijk lang.
Affimed, vorige week mijn laatste 25% verkocht. Dit gokje in het biotech casino leverde mij een min op van $7,675. Had dat geld vorige week ook weer terug gebracht naar ASML. Dat is sindsdien heel fors gestegen. Het was weer eens het bekende biotech uitje ;-)

Ben blij er zelf niet in te zijn gestapt. Arwh Wel paar weken terug. Tja, shit happens, niet getreurd, de rest is redelijk. Vandaag mooie dag, gemiddeld +1,5% . Vanaf 1 januari al 5.4%. Ik heb zo en zo een heel mooi bedrag aan een goed doel geschonken. Dat doe ik elk jaar.
Gezien ook de tik die Nkarta vandaag krijgt (- 10,7%) en de wederopstanding van Fate (+27%) lijkt het er op dat NK cellen geheel uit de gratie zijn. Een kwestie van kortzichtigheid ?
