Het jaar van de waarheid

Is toch inmiddels behoorlijk bloedbad.
www.fool.com/investing/general/2015/1... No. 3: Doubts emerge for NASH treatment Nonalcoholic steatohepatitis (NASH) is a fast-growing cause of liver failure that is estimated to occur in 5% of patients and it could become the leading cause of liver transplant by 2020. In a bid to keep that from happening, Intercept Pharmaceuticals (NASDAQ:ICPT) is developing obeticholic acid. Last year, positive mid-stage findings resulted in Intercept Pharmaceuticals kicking off a phase 3 study in the U.S. with a scheduled interim look in 2017, but enthusiasm for NASH therapy has faded since competitor Genfit reported in March that its NASH drug failed in phase 2 studies and Intercept Pharmaceuticals reported in October that a phase 2 study of obeticholic acid in Japan failed to improve patient's liver disease by two points on a standard liver disease measure. Because these trial failures raise concern that obeticholic acid may not be as effective as previously thought, shares in Intercept Pharmaceuticals have fallen to less than $150 from a peak of $480 earlier this year. That's a lot of pain for investors to endure, but it's not all bleak for Intercept Pharmaceuticals. The company has already filed for approval of obeticholic acid in another much rarer form of liver disease and a decision from the FDA in that indication is expected in February. Additionally, there's still a chance that obeticholic acid posts better results in the larger ongoing U.S. phase 3 study than it did in this smaller Japanese phase 2 trial.

The FDA will speed up its review of Shire’s ($SHPG) experimental midstage fatty liver disease drug volixibat as the battle to be first to market in this emerging disease area heats up. The Irish biopharma has been given the track designation from the U.S. regulator for SHP626 (also known as volixibat) for nonalcoholic steatohepatitis (NASH) with liver fibrosis. This condition, which is estimated to affect millions of Americans, is directly related to the rise of obesity and type II diabetes, and can lead to liver cirrhosis, cancer and death. “Shire’s development plan for SHP626 is designed to address the unmet need in the treatment of adult patients who have NASH with liver fibrosis,” said Philip Vickers, head of R&D, Shire. “This fast track designation is further recognition of the critical need to develop new, effective therapeutic options for patients with this serious condition.” There are currently no approved meds in this area, but analysts believe--as with the hep C drugs from Gilead ($GILD)--a highly efficacious treatment that could cure patients of the condition could be a major blockbuster contender, with predictions being as high as $10 billion at peak per med. Shire is racing Gilead, Intercept Pharmaceuticals ($ICPT), Genfit and others in what could eventually be a $40 billion market, although most of its rivals are further up the path than the biopharma in development terms. Intercept appears to be the nearest to market after gaining in May an FDA approval for Ocaliva in a separate liver disease license known as primary biliary cirrhosis (PBC)--in what is being seen as a precursor to a NASH license. The biotech has however come up against problems as last year it flubbed a Phase II NASH test, but it has shown promise in other trials. A year ago, Intercept began enrollment in a new global study for its NASH candidate designed to show if it can reduce liver scarring and prevent death and end-stage liver disease. Final data won't be ready until 2021, but Intercept has scheduled an interim analysis in 2017 that will evaluate how well its drug is doing at resolving NASH and reducing scars in about 1,400 patients. It has said it plans to submit those results to the FDA for a shot at early approval.Meanwhile Genfit, likely a second-place contender, has told FierceBiotech that it expects its drug to be approved by the FDA in 2019. Shire and Gilead are a little further down the ladder. Shire’s candidate is a once daily pill that works as an inhibitor of the apical sodium dependent bile acid transporter--a protein which is primarily responsible for recycling bile acids from the intestine to the liver. It’s been in Phase I but is now set to move into midstage, placebo-controlled studies. The drug was originally licensed from Lumena Pharmaceuticals.
$40 Billion Opportunity: 3 Biotechs Poised to Dominate The NASH market should be huge. Gilead Sciences, Intercept, and Allergan stand ready to profit. Keith Speights (TMFFishBiz) Oct 31, 2016 at 10:03AM Nonalcoholic steatohepatitis (NASH) has been called "the next hepatitis C" by some. Both NASH and hepatitis C are liver diseases -- and both represent huge markets for drugmakers working on cures. While the hepatitis C treatment landscape has filled out over the past few years with multiple effective options for patients, the NASH market is still in its infancy -- but the potential is huge. It's estimated that NASH affects 2% to 5% of Americans, and analysts have projected that the global NASH treatment market could reach $40 billion by 2025. Three biotechs appear to be poised to capture large shares of the NASH market: Gilead Sciences (NASDAQ:GILD), Intercept Pharmaceuticals (NASDAQ:ICPT), and Allergan (NYSE:AGN).
Although Gilead has shied away from outright acquisitions, it has engaged companies such as Galapagos NV with collaboration deals that involve little upfront but could be worth much more if they pan out in the clinic. Through the Galapagos partnership, Gilead has advanced filgotinib into late-stage studies for rheumatoid arthritis, Crohn's disease, and ulcerative colitis. If successful, peak annual sales of the drug could reach $3 billion, despite risking just $725 million upfront.Further ahead, the company has three separate mid-stage candidates that comprise what is arguably the best shot at becoming the first effective treatment for non-alcoholic steatohepatitis. This underserved population represents a market estimated at about $15 billion annually. In the near term, America's largest pharmacy benefit manager, which once shunned Gilead's best-selling hepatitis C antiviral to date, has recently agreed to add the drug to its 2017 formulary. These lights ahead won't send Gilead Sciences investors laughing all the way to the bank, but the company's future isn't nearly as grim as its ultra-low valuation suggests. Truckloads of free cash flow have to go somewhere, and I'd much rather see it spent repurchasing beaten-down stock than chasing overpriced biotechs.www.fool.com/investing/2016/12/20/gil...

Mijn mening: waardeloze resultaten.
