This trial has also been used to provide safety data for the BLA filing for leronlimab as a combination therapy. In view of the high responder rate at the increased dosage levels, coupled with the newly developed CCR5 occupancy test, we filed a pivotal trial protocol with the FDA for leronlimab as a monotherapy in May 2019". Folks, per latest CYDY SEC financial report filed 10/9/20, CYDY is looking at approval of Leronlimab for COVID-19 illness based on a successful P3 CD12 trial (EUA is no longer in play at this stage IMHO). If interim analysis of 50% enrolled subjects is statistically significant and CD12 trial is stopped for efficacy this coming week, Leronlimab can be the first treatment to get conditional or accelerated approval for COVID-19 illness. Question will be how broad will label be, just severe illness or moderate-severe illness(or mild-severe). We will know by end of this coming week. Per CYDY on page 34 first paragraph: "As described above, the Company is currently conducting a Phase 2b/3 clinical trial for 390severe-to-critically ill COVID-19 patients. If results from this trial indicates statistically significant clinical outcomes for the COVID-19 patients to sufficiently meet the primary and secondary endpoints for the trials, the Company expects to seek FDA approval."

Hoe moeten we rekenen m.b.t. CD12? Hier doet iemand het voor. Ook een zinnige reactie erop. Simplistic math. 45 deaths total of 195. Forget Pvalues for a moment because we’re not trying to get approved for heart burn or high blood pressure where there are tons of approved drugs already. keep in mind leronlimab arm has twice as many Patients as it’s a 2-1 ratio Nadar wanted to be able to help the majority in the trial. I’ve researched many large studies finding that avg of 30-35% avg mortality in severe critical which would put placebo at around 21 deaths in Placebo and 24 in leronlimab. I’m not a Pvalue expert but if you double the 21 you now have a 1-1 comparison of 24 leronlimab 42 placebo this would be a mortality rate reduction of 57% reduction in mortality. Head of the FDA Steven Hahn quoted saying 35% mortality reduction is a dream number. That 35% reduction would be shown with 27 deaths in leronlimab and 18 in placebo. Which 18/65 in placebo group would be 27% mortality rate. I don’t believe placebo performed that well. If anything I believe most of the trial patients would have been in the worst shape desperate for an experimental drug when whatever they were taking was failing. If that’s the case the placebo mortality might be as high as 37% which would be 24 deaths in Placebo and 21 in leronlimab. This would have a pvalue low enough for approval. Any of the other scenarios should still get EUA in my opinion. There aren’t to many scenarios where is see placebo out performing leronlimab given the fact when taken early enough in m/m reduced SAE 65% and avg mortality rate in EIND patients I believe was around 18%. If you use the 18% it would be 23 patients in leronlimab vs 22 in placebo once again Pvalue met and approval using all the data we currently have.The % reduction alone doesn’t tell you enough. If treatment had 1 death and placebo had 2 deaths, that would be a 50% reduction but the chances of that happening randomly are very high unless you tested 1000s of patients. You need to have enough patients in the study to ensure the any reductions in mortality are real and not by chance. The wider the gap the less patients you need to be confident that it was not a random event. Seems to me IF Leronimab performed as well as you suggest the fda would have halted the trial

CytoDyn is facing a key catalyst within days as regards mortality in its severe/critical COVID-19 trial. CytoDyn's rush to assure adequate supplies of leronlimab, a molecule which has yet to be approved anywhere for anything, presents jumbo challenges. I took on a role as chronicler of the tales of CytoDyn (OTCQB:CYDY) with enthusiasm over recent months. I wrote articles, participated in comment streams, purchased and sold shares (on the long side only) and generally edified myself as I hibernated in COVID-19 induced isolation. After a longish period as a strong CytoDyn skeptic, I had moved from a bearish view to a neutral stance and then cautious optimism. In the first bullish article, "CytoDyn's COVID-19 Game Changer", I tried to draw a nuanced portrait of CytoDyn's risks compared to its prospects. Such article featured a bullet point that "CytoDyn's leronlimab is shaping up as a prime contender to be a preferred COVID-19 therapeutic". I would be hard pressed to make that statement today. This article reviews recent developments that have diminished CytoDyn's prospects. Early shareholder optimism around leronlimab in treatment of mild-to-moderate COVID-19 is fading CytoDyn was early in advancing its leronlimab as a COVID-19 therapy. Its EIND program success was highly suggestive that leronlimab would prove out as effective in treating COVID-19. More importantly, CytoDyn mapped out a phase 2 clinical trial ("CD10") for mild-to-moderate patients and a phase 2/3 trial ("CD12") for severe/critical patients. CD10 was a trial designed for 75 patients, although 86 were actually treated. It required 14 days of weekly treatments with 700 mg leronlimab or placebo with study participants randomized 2:1. The primary outcome measure was: CytoDyn's CEO Nader Pourhassan (hereafter "NP") encouraged shareholders to expect quick readouts of the data from CD10. As it turned out, CytoDyn began reporting driblets in various press releases. In what has become a tedious stylistic trope, the headlines for the releases have been overblown with the content often unremarkable. This was particularly true of CytoDyn's first such release in the early hours of 7/21/20, subsequently corrected a few hours later, as discussed in "CytoDyn's COVID-19 Trial Tribulations". The bottom line is that CD10 missed its primary endpoint. In a subsequent 8/11/20 press release, CytoDyn announced that CD10 achieved statistically significant results in a key secondary outcome measure, National Early Warning Score 2 scale (NEWS2). In its 8/12/20 conference call CytoDyn announced that it had applied to the FDA for an emergency use authorization [EUA] based on the "fantastic" NEWS2 news. As far as I have been able to tell, this was the situation that shareholders thought was applicable until NP's 9/23/20 interview with Dr. Been. In this interview NP explains that CytoDyn ran the idea of applying for an EUA by the FDA but elected not to when it became clear that it was unlikely to succeed. Leronlimab in treatment of severe/critical COVID-19 presents a catalyst expected in days As for CD12, it is a bigger phase 2b/3 trial with a longer treatment period. Full enrollment requires 390 patients, although as a phase 2b/3 trial there will be a preliminary read out when half of the participants have been treated. The treatment is the same as CD10 but extended to include weekly doses of 700 mg leronlimab or placebo for 28 days, randomized 2:1. The action for CytoDyn at the current point is all about CD12. During CytoDyn's 8/12/20 conference call, NP advised that 175 patients had been enrolled, just 20 less than the 195 required for the interim analysis. By 9/23/20 NP advised during his Dr. Been interview that enrollment stood at ~215. He rightly noted the criticality of completing the trial avowing an intent to do so by the close of 2020 with data submitted to the FDA. Moving on to CytoDyn's 9/30/20 shareholder's conference, CytoDyn issued the following slide relevant to CD12 (discussed 24:05-25:20/1:16:01). During the discussion on CD12, NP emphasized how information already known from the EIND and from the safety look following 100 patients treated derisked the situation. He posited that the interim analysis presents the potential first time a COVID-19 trial hits a primary endpoint of mortality.

Ik heb niet onder stoelen banken gestoken dat ik kritisch ben over Cydy en de Covid-kansen, noch over Naders manier van leidinggeven. Ik heb dit allemaal zien aankomen en daarom ook al afgebouwd. Wat ik wel vreemd vind van jouw kant is dat je het middel tegen HIV blijkbaar ook niet meer op waarde weet te schatten. Ik kan zo een post boven water halen van jouw kant dat je daar miljarden van verwachtte. Leronlimab als middel tegen Covid-19 zou zelfs volledig ondergeschikt zijn aan Leronlimab als middel tegen HIV. Los van de financiële perikelen, heb je ook geen vertrouwen meer in de op handen zijnde BLA, 700 mg / HIV?
Hier een iets meer genuanceerde reactie op de naderende ontluiking van de DSMC berichtgeving dinsdag 20 okt. a.s.: Looks like the path forward is either complete the 390 patient trial, something no one else has done successfully with a safe and significantly effective drug, or complete it changing the end points or, if the 60 eIND results and CD010 mild/moderate results hold, EUA with close follow up. Seems like the latter two are more likely because unless something game changing happened, Cytodyn would not have received any DSMC results. Shorts need to continue to control the price to protect their 24 million of short interest. But they can't control the science which inexorably has been going against them.Summary: We did not get shut down on the Phase 3. 2 possible scenarios: 1. No EUA, but continue the trial and will review at 395. (this is good news!) 2. Recommendation for EUA submission and CYDY is currently submitting and will reveal this on Tuesday.

So the glass is about half full. I was wondering about how the 11 big pharma companies will think about this? Will anyone of them be interested enough to get into action? Also, will Cytodyn management be humbled enough to get into one agreement? Or, will they be so encouraged that they will continue to go it along so that they can get a better deal later? I am holding my positions but I am not expecting huge short-term price jumps. Stay LONG!

Ik dacht dat je een GAK had van 0,4!? Als je nu het grootste gedeelte hebt verkocht krijg je nooit meer zo'n GAK - tenminste...als het (een beetje) goed is. Ik denk dat ze dinsdag gaan voor EUA, maar dan moeten ze wel snel kunnen aantonen dat ze een aanzienlijk hoeveelheid Leronlimab kunnen produceren binnen korte tijd. Gezien de contacten met Samsung en die bekende distributeur moet dat wel kunnen. Verder als de BLA op de rails staat leidt dat tot meer welwillendheid aan de financiële kant. Mijn mening kort: die neerwaartse spiraal kan stopgezet als ze maar iets voor elkaar krijgen. Nu hebben ze concreet nog niets. Als dat blijft zal geld ophalen moeilijk worden. Haha. Ik kan zo 10 posts uit dit forum boven water halen waaruit blijkt dat je op je bek ging, overdreef, het fout had, of inconsistent was t.a.v. Cydy en de ontwikkelingen. Je verweer is dat van een kind. Scheer je weg van dit forum nu je verkocht hebt (wat je nooit zou doen, want de wetenschap is superieur.) En er gaat ook geen volledige negatieve spiraal plaatsvinden naar onder de 1, let maar op!

De enige die hier het aandeel uitkotst ben jij. Jij verkoopt. Typisch voor een kind: je doet zelf wat je anderen verwijt. Je doet alsof je beter bent dan de rest terwijl je gewoon je zakken (probeert) te vullen. Er is helemaal niets bijzonders aan Leronlimab behalve dat het de markt zou kunnen halen voor enkele indicaties en doelgroepen. Er is al helemaal geen samenzwering van BP.
