Een onafhankelijke commissie van wetenschappers heeft nog geen tussentijdse analyse kunnen uitvoeren die moet uitwijzen of het middel effect heeft, zo meldde het concern in een kwartaalupdate. De tussentijdse analyse is pas mogelijk als minstens 32 deelnemers aan een grootschalig onderzoek naar het vaccin besmet zijn geraakt met het nieuwe coronavirus. Topman Albert Bourla stelde onlangs in een open brief dat Pfizer mogelijk eind november al om versnelde goedkeuring voor het vaccin zal vragen bij de Amerikaanse toezichthouder FDA. Dat zal alleen gebeuren als het middel veilig wordt geacht en er bewijs is dat het vaccin effect heeft, zo schreef hij. Pfizer had ook opbeurend nieuws over het vaccin. De werving van deelnemers aan zijn zogeheten Fase 3-onderzoek naar het coronavaccin is bijna rond. Inmiddels doen ruim 42.000 mensen mee aan klinische tests met het vaccin, terwijl het Amerikaanse concern zich ten doel heeft gesteld 44.000 deelnemers te werven. Pfizer meldde in zijn kwartaalupdate dat 36.000 mensen een tweede dosis hebben gekregen van het middel, dat samen met Duitse BioNTech wordt ontwikkeld. Pfizer gaf de update samen met zijn cijfers over het derde kwartaal. Daarin daalden de opbrengsten met 4 procent tot 12,1 miljard dollar. Dat kwam onder andere doordat te midden van de coronapandemie minder Amerikanen op doktersbezoek gingen, dus er ook minder medicijnen voorgeschreven werden. Daarnaast was de vraag naar medicijnen in China lager door de virusuitbraak.

Dit bleek donderdag uit de kwartaalupdate van het Amerikaanse biotechbedrijf. In een jaar tijd ging de omzet van Moderna van 17 miljoen naar 158 miljoen dollar. Deze stijging is volgens Moderna te danken aan een stijging van de subsidie-inkomsten, voornamelijk door de BARDA-overeenkomst voor de ontwikkeling van een coronavaccin. Met deze omzet overtrof Moderna de marktverwachtingen ruimschoots. Netto boekte Moderna nog altijd een verlies van 234 miljoen dollar. Een jaar eerder was dit een verlies 123 miljoen dollar. Per aandeel bedroeg het verlies 0,59 dollar. Moderna sloot het derde kwartaal met bijna 4 miljard dollar in kas. De cashburn bedroeg 44,1 miljoen dollar in de eerste negen maanden van 2020. CureVac Posts Positive Early Data For Its Covid-19 Vaccine Candidate 11/02/2020 Recent hot IPO CureVac (CVAC) on Monday posted positive preliminary data on a Phase 1 clinical trial for its Covid-19 candidate. CVAC stock seesawed on the news, closing down. The German company is a pioneer in messenger RNA technology. It is among a number of companies working on mRNA Covid-19 vaccine candidates. The gene-based vaccines are designed to direct the body's cells to create a protein similar to the spike protein found in SARS-CoV-2, triggering the body's natural immune response. Covid-19 is caused by the SARS-CoV-2 virus. On Monday, CureVac said its Phase 1 tests showed the vaccine was generally well tolerated and induced strong neutralizing antibody responses. The company said it plans a detailed overview of the trial results and publication in a scientific journal in the coming weeks. In the Phase 1 study, more than 250 healthy adults ages 18 to 60 received two vaccinations 29 days apart. This included as many as 10 who had previously tested positive for Covid-19. Those efforts, too, are further behind the leaders. Last month, Sanofi and Translate Bio posted positive pre-clinical trial results for their vaccine candidate and said Phase 1/2 clinical trials would start this month. For its part, CureVac is amid a Phase 2a clinical trial for a Covid-19 vaccine in Peru and Panama. CVAC Stock Soared Fivefold First Two Days On Market On the stock market today, CVAC stock rose as much as 4.6% early on the news. But later in the session, shares reversed and fell as much as 4.7%. At the close, CVAC stock fell 3.5% to 47.08. CureVac made its U.S. initial public offering on Aug. 14. The company priced CVAC stock at 16 but shares closed at 44 that day. The next day, it touched a record high of 85. Since that initial burst, CVAC stock generally has trended lower. In late September, it touched 43, its lowest price since its first trading day. Still, CVAC stock carries an Accumulation/Distribution Rating of A+, which indicates heavy institutional buying.

Dit meldden de twee farmabedrijven maandag in een interimanalyse van een Fase 3 onderzoeksstudie. Aan het onderzoek doen 43.538 proefpersonen mee en er kwamen tot nog toe geen serieuze problemen aan het licht betreffende de veiligheid. De werkzaamheid werd gemeten onder proefpersonen die twee doses van het vaccin kregen en deelnemers met een placebo. Volgens de bedrijven werd bescherming tegen het vaccin bereikt 28 dagen na de eerste toediening. Pfizer en BioNTech benadrukken dat de definitieve werkzaamheid nog kan afwijken, terwijl de studie wordt voortgezet. Pfizer en BioNTech verwachten binnen enkele weken een noodvergunning, een zogeheten Submission for Emergency Use Authorization (EUA), aan te vragen bij de U.S. Food and Drug Administration. Hoe werkt het mRNA-vaccin? BNT162b1 is een zogenoemd mRNA-vaccin en werkt net ietsje anders dan de traditionele vaccins. Traditionele vaccins bevatten vaak een verzwakte of dode ziekteverwekker. Door die in het lichaam in te brengen, kan het immuunsysteem antistoffen tegen die ziekteverwekker aan gaan maken. En wanneer je lijf dan eens met een levende of sterkere versie van zo’n ziekteverwekker geconfronteerd wordt, is het al voorbereid en kan het direct in de aanval, waardoor de ziekteverwekker je niet ziek kan maken of de ziekte mild verloopt. mRNA-vaccins zijn vrij nieuw en werken anders. Hierbij wordt RNA – eigenlijk de instructies voor het maken van een stukje van de ziekteverwekker – in het lichaam ingespoten. Dit RNA nestelt zich in je cellen, waarna deze de instructies op gaan volgen en dat stukje van de ziekteverwekker – een eiwit – gaan aanmaken en het immuunsysteem er antistoffen tegen kan genereren. Op zich dienen vaccins natuurlijk veel langer getest te worden. Maar vaccins zijn wel relatief simpele medicijnen. Ze werken direct met het immuun-systeem - in dit geval door mRNA van de ziekteverwekkers te injecteren. In vergelijking tot bijv. filgotinib, dat werkt op JAK, is een vaccin veel eenvoudiger. JAK maakt onderdeel uit van het JAK-STAT proces dat een rol speelt bij alles, van immuniteit tot celdeling en kan door tientallen verschillende stofjes geactiveerd worden. Met een dergelijk gecompliceerd medicijn heb je een veel grotere kans op bijwerkingen dan met een simpel vaccin.

The company’s effort to deliver relief to pandemic-weary populations will revolve around refrigerated storage sites at two of the company’s final assembly centers—the Kalamazoo facility and another in Puurs, Belgium—and rely on dozens of cargo-jet flights and hundreds of truck trips every day. Distribution centers in Pleasant Prairie, Wis., and in Karlsruhe, Germany, have been outfitted for extra storage capacity. Pfizer so far has spent about $2 billion on developing the vaccine and setting up the distribution network. The U.S. government placed an initial order for 100 million doses, with the option to purchase 500 million additional doses. The EU ordered 200 million doses with an option for another 100 million. Japan ordered 120 million doses and the U.K. 30 million. Countries in South America and in the Asia-Pacific region also have placed significant orders. In a typical vaccination campaign, pharmaceutical companies would wait until their product is approved before buying raw materials, establishing manufacturing lines and setting up supply chains to ship a vaccine. Pfizer Chief Executive Albert Bourla said that the company began setting the groundwork for its supply chain in March, when it kicked off its vaccine development. “Ensuring over a billion people globally have access to our potential vaccine is as critical as developing the vaccine itself,” he said. Pfizer says it is preparing for distribution in case the vaccine wins authorization, with hundreds of thousands of doses already in the company’s warehouses in the U.S. and Europe. Cool Box To make sure its Covid-19 vaccine doses arrive at hospitals and clinics frozen and potent, Pfizer created its own container to ship them. The temperature-controlled container can store between 1,000 and 5,000 doses for 10 days at minus 70 degrees Celsius before requiring re-icing.

Source: Pfizer To keep the vaccines safe in transit and to move them fast, Pfizer designed a new reusable container that can keep the vaccine at ultracold temperatures for up to 10 days and hold between 1,000 and 5,000 doses. The suitcase-sized boxes, which are packed with dry ice and tracked by GPS, will enable Pfizer to avoid the larger, temperature-controlling containers used in transportation, giving it more flexibility to ship the vaccines faster since planes and trucks won’t have to wait for the standard refrigerated metal boxes. Pfizer expects to load those boxes on a combined 24 trucks a day from Kalamazoo and Puurs that will move roughly 7.6 million doses daily to airports. The company plans to take cargo space on an average of 20 flights a day on planes operated by FedEx Corp. , United Parcel Service Inc. and DHL International GmbH to fly the vaccines as close as possible to vaccination centers, ranging from big medical facilities to far-flung hospitals. The air carriers are also in line to handle the next leg of the vaccine’s journey, trucking the doses to sites close to where they will be administered. Total delivery time, from distribution center to point of use, is expected to be an average of three days, the company said. Cargo airlines are scrambling to arrange scores of extra flights to move the vaccines. They could hit distribution channels at the height of the peak season for shipping goods ahead of the year-end holidays, squeezing expedited shipping capacity. Unlike traditional vaccine rollouts, Pfizer plans to bypass distribution wholesalers, including McKesson Corp. , which has been tapped by the U.S. government to distribute Covid-19 vaccines through the federal Operation Warp Speed program. “For the most part, we are not going to be building inventory,” Ms. Alcorn said. In het derde kwartaal boekte BioNTech een omzet van 67,5 miljoen euro. Dat was flink meer dan de 28,7 miljoen euro een jaar eerder.
