Eyeing 'a bit of a bubble,' Lilly shies away from biotech buyouts Biotech's ongoing boom has inflated valuations for upstart drugmakers, and Eli Lilly ($LLY) CEO John Lechleiter said his company is wary of getting involved in any high-dollar bidding wars to bring in new treatments. Speaking to analysts following Lilly's second-quarter earnings presentation, Lechleiter said "there's a bit of a bubble" at play in the biotech world, jacking up prices for promising assets in development by small and mid-size companies. And Lilly, in need of a sales boost, is less than eager to risk overpaying for new drugs, he said. Lechleiter's comments echo a similar sentiment from Roche ($RHHBY) CEO Severin Schwan, who told CNBC on Thursday that the company has turned down a number of deals because the market price for new therapies has simply gotten too high. Interestingly, those rising concerns among Big Pharma aren't broadly shared among the industry's middle class. Celgene ($CELG), a sizable biotech competing with global giants, has been repeatedly willing to cut big checks for new assets, this year signing a $1 billion deal with Juno Therapeutics ($JUNO) in oncology and paying $7.2 billion for Receptos ($RCPT) and its anti-inflammatory drugs. And Alexion Pharmaceuticals ($ALXN), another Big Biotech, shelled out $8.4 billion for fellow rare disease specialist Synageva BioPharma in May. Such big-budget bets reflect a greater appetite for risk, as, especially in Celgene's case, there are major binary events on the calendar that could make those acquisitions look either prescient or wrongheaded. And to Lechleiter's point, investors have taken a very bullish stance on such gambles; Celgene's shares are up roughly 24% on the year, while Alexion's have risen about 11%. But Lilly, which finally delivered sales growth in Q2 after 6 consecutive quarterly slumps, may not be in a position to be choosey. The company's late-stage pipeline is full of drugs that, if approved, would compete in crowded markets, whether in diabetes, inflammation or oncology. De verkopen van het MS-middel Tecfidera vielen in het tweede kwartaal tegen en de groep verlaagt ook de winstprognose voor het volledige boekjaar. Anailsten stippen aan dat Tecfidera flink wat concurrentie krijgt van Novartis' middel Gilenya. Key data for the last few oral antirheumatics Date July 24, 2015 Remember when in the early part of the decade it was widely held that cheap, oral antirheumatic drugs would shortly replace expensive, injectable anti-TNF agents for rheumatoid arthritis? Well, that seems rather rash now. Pfizer’s Jak inhibitor Xeljanz, the lead agent in this class, did make it to the market in 2012 but has failed to live up to expectations, while its close competitors, including AstraZeneca and Rigel's syk inhibitor fostamatinib, have been dropped from development. With the once-rich pipeline quietly whittled down to perhaps six oral agents in late development today, it is now the turn of Lilly's baricitinib to show whether an oral agent can still reshape the RA space. Results are due from trials of several other agents too, but baricitinib appears to be the most important. Consensus sales forecasts for it stand at $650m in 2020, according to EvaluatePharma. The most important trials of baricitinib, a Jak-1/2 inhibitor, are two phase III studies known as RA-Begin and RA-Beam. Steady newsflow RA-Beam enrolled 1,280 patients with an inadequate response to methotrexate and tested 4mg of baricitinib versus AbbVie’s Humira as well as placebo. RA-Begin tests two doses of baricitinibversus placebo in 550 patients with limited or no treatment with methotrexate, and who are naïve to other conventional or biological disease-modifying antirheumatic drugs (DMARDs). Lilly has already reported positive results for two other phase III trials, RA-Build and RA-Beacon, and replication of these data should ensure baricitinib's place in the market. That place may not go unchallenged. An important competitor, AbbVie, will shortly have to make a decision on whether to opt into a pre-negotiated licensing agreement with Galapagos for the Belgian company’s selective Jak-1 inhibitor, filgotinib. AbbVie paid $150m in 2012 to secure the option, with a further $200m payable on exercise and further milestones of up to $1bn and tiered double-digit royalties. That decision is now looming and will be based on 24-week data from two phase II studies expected in the next few weeks (Galapagos moves towards RA deal but a dark horse remains on the horizon, April 15, 2015). Under control Although the full Phase III data on baricitinib would be an important consideration for AbbVie, one other factor within its control is the outcome of a phase II study of an internally developed Jak-1 inhibitor, ABT-494. This trial has recently been completed, so presumably results are already available. Analysts covering Galapagos have assumed that AbbVie will exercise its option, though competition in the Jak inhibitor area is fierce. This was presumably behind J&J’s decision last December to discontinue a joint project with Astellas for the only other Jak inhibitor in development, ASP015K. J&J had held ex-Japan rights. Astellas is continuing Japanese development, with two phase III studies under way and due to render results in March 2017. There are only two other oral DMARDs in mid to late development: Celgene’s BTK inhibitor, CC-292, and Pfizer’s selective glucocorticoid receptor modulator, PF-04171327. Bron De Tijd 27/07/15 Minister van Volksgezondheid Maggie De Block (Open VLD) heeft een reeks maatregelen afgesproken met de farmaceutische industrie. Hierdoor worden geneesmiddelen goedkoper en zullen ze ook sneller op de markt komen. De overheid en de farma-industrie gaan een reeks maatregelen doorvoeren waardoor geneesmiddelen in totaal jaarlijkse zo'n 24 miljoen euro goedkoper worden. Dat bedrag zal met de jaren oplopen. In 2018 zullen patiënten al 60 miljoen euro kunnen besparen op hun geneesmiddelen. Minister van Volksgezondheid Maggie De Block kwam na maanden onderhandelen tot een akkoord met de farma-industrie. De twee partijen ondertekenden deze morgen een 'toekomstpact'. Geneesmiddelen worden hierdoor niet alleen goedkoper, bovendien zullen ze sneller worden goedgekeurd en op de markt komen. Zodat patiënten die nood hebben aan een nieuw geneesmiddel, er sneller toegang krijgen tot krijgen. Minister De Block gaat ervan uit dat medicijnen minstens twee maanden vroeger beschikbaar zullen zijn. Dat is een grote verbetering, want vandaag duurt het in België vaak meer dan een jaar voor dat de terugbetaling van een geneesmiddel wordt afgesproken en het middel vervolgens op de markt kan komen. In andere landen gaat die procedure doorgaans sneller. De overheid belooft de farmabedrijven in ruil voor de tegemoetkomingen een stabiel klimaat, zodat ze verder kunnen groeien. België is het land waar de meeste klinische proeven gebeuren per inwoner en de farmaproducenten drongen er dan ook op aan dit zo te kunnen houden. 'De overheid heeft een stabiel, voorspelbaar en betrouwbaar budgettaire situatie gecreëerd waarbinnen de farmaceutische sector zijn onderzoek en ontwikkeling beter kan plannen', meldt minister De Block in een persbericht. Verder blijkt uit de 'toekomstpact' dat de overheid dankzij de nieuwe afspraken met de farmasector zelf jaarlijks 226 miljoen euro zal besparen. Via die besparingen kan de overheid aan de industrie een belastingverlaging van 13 procent op de omzet toestaan.
Bron De Tijd 28/07/15 UCB De ontgoochelende resultaten in de derde en laatste testfase zijn een tegenvaller voor de Belgische biofarmaciegroep én voor lupuspatiënten die slechts weinig nieuwe middelen op de markt zien komen. De decennialange zoektocht naar een wondermedicijn tegen lupus, een ziekte van het immuniteitssysteem die vooral jonge vrouwen treft, zal nog wat langer duren. De resultaten van de derde en laatste klinische testfase voor epratuzumab, het potentiële UCB-middel tegen lupus, vallen tegen. 'De resultaten bij patiënten die epratuzumab samen met de standaardtherapie toegediend kregen, waren niet significant beter dan bij mensen die een placebo samen met standaardtherapie kregen', zegt de biofarmaciegroep in een mededeling. De resultaten zijn een ernstige tegenvaller voor UCB. Epratuzumab maakt deel uit van het 'BER'-trio dat de komende jaren de groeimotor moet vormen voor UCB. De andere twee zijn brivaracetam tegen epilepsie en romosozumab tegen osteoporose. Beleggers reageren ontgoocheld. Het aandeel UCB zakt 5 procent of 3,3 euro per aandeel. Dat is ook logisch: de meeste analisten zetten nu in epratuzumab in hun waarderingsmodel op nul. Bij beushuis Credit Suisse, bijvoorbeeld, dichten ze vóór de resultaten het middel nog 2,6 euro per aandeel UCB toe. Zowel analisten als het management hebben echter steeds gewaarschuwd dat het lupusmedicijn het meest riskante onderzoek is. 'Het is een product waar we veel risico mee nemen. Iedereen zoekt bij wijze van spreken naar een geneesmiddel tegen lupus, maar niemand vindt iets', zei CEO Jean-Christophe Tellier op de aandeelhoudersvergadering eind april. Benlysta, het lupusmiddel van GSK, was in 2011 het eerste nieuwe lupusmedicijn in 56 jaar. Maar de verkoopscijfers vallen tot nog toe tegen. In 2014 bleef de verkoop steken op 155 miljoen pond (218 miljoen euro).
