    We’re glad to see Arrowhead continue to put additional development candidates into the clinic with the start of the Phase 1/2a trials for ARO-RAGE and ARO-MUC5AC. The company learned a number of things from the ARO-ENaC program that should be transferrable to these new pulmonary programs, thus increasing the probability for a clinically successful outcome. While there haven’t been too many data readouts recently, we anticipate 12 clinical readouts between now and the end of 2023 when including both wholly-owned and partnered assets. Thus, investors should expect a lot of news flow over the next 12-15 months. We have made no changes to our model and our valuation remains at $92 per share.

    Beurzen reageren positief op VS-inflatie
    De Europese beurzen reageren positief op het inflatierapport. De Bel20 zit plots op een winst van 1 procent. Ook de Amerikaanse futures staan duidelijk op winst.

    Amerikaanse inflatie daalt meer dan verwacht
    De Amerikaanse inflatie is in juli gedaald naar 8,5 procent, tegenover 9,1 procent in juni. De kerninflatie bleef stabiel op 5,9 procent, terwijl een stijging naar 6,1 procent was voorspeld.

    De lager dan verwachte inflatiecijfers verminderen de kans dat de centrale bank (Fed) in september de rente opnieuw met 75 basispunten verhoogt.

    Ik zag plots de €/$ stevig wijzigen in dollarbeleggers hun nadeel, hierboven de reden.

    Toch raar dat er volop wordt ingezet op indicaties die niet levensbedreigend zijn. Vraag me af of de gen-jongens (lees: big pharma die ze ruggensteunen?) niet een dikke vinger in de pap hebben bij de FDA...

    Don't know how or why Verve is doing gene editing for targets that are already being addressed by other modalities. Both PSCK9 and ANGPTL3 are not only being knocked-down with RNAi, but the potency is more than enough. Shocked that the medical community is not standing up to this.     Conclusion:

    No gene editing technique is perfect in all situations. The optimal gene-editing modality depends on the specific application, biological context, and potential trade-offs. That said, CRISPR-Cas9 nuclease treatment can result in dozens of uncontrolled indel outcomes in the effort to disrupt genes and does not result in the precise conversion of one DNA sequence to a specific sequence; its double-strand breaks also can cause unintended genetic consequences. Base editors make A-to-G, G-to-A, C-to-T, and T-to-C edits precisely and result in fewer indel or other editing outcomes but can cause bystander edits that may or may not have biological consequences. Prime editing seems to be the most versatile of the gene-editing technologies, resulting in few undesired outcomes at the expense of efficiency.

    Understanding the risks and benefits associated with each gene-editing method is important. A careful examination of all the DNA sequence outcomes reveals how different gene-editing technologies can change target DNA, informing researchers of the best use cases for each. These breakthrough technologies aim to correct DNA mutations associated with thousands of genetic diseases––permanently. They are not treating or controlling symptoms of disease. Medicine has entered a new era.

    Geen woord over alternatieven uiteraard, rnai zal wel als ouderwets gezien worden.
    Gene editing blijft sexy en alle fouten die nu nog optreden zullen wel als kinderziekten bestempeld worden.
    Opgeteld bij het feit dat gene editors ook 'gewoon' inzetten op grote (blockbuster) indicaties die niet levensbedreigend zijn én het uitblijven van grote (ethische) bezwaren uit de markt hieromtrent, maakt mij toch wat voorzichtiger. Ik weet wel dat de FDA een tijd geleden heeft gesteld dat gene editing pas mag als er geen alternatief bestaat (kan de bron zo snel niet terugvinden), maar veel vertrouwen in dat orgaan heb ik niet als het aankomt op rechtschapenheid en authenticiteit.
    Er zullen nog veel meer gene-editing mislukkingen aan het licht moeten komen om de markt weg te krijgen bij de crisprs, beams en intellia's richting ARWR en ALNY.

    Hoe denken jullie hierover?

    Op twitter zie ik antwoorden passeren Hulshof maar zelf kan ik weinig toevoegen.

    Wat anders Goldman Sachs verlaagde koersdoel destijds van $92 naar in de 60.
    Blijkbaar kochten ze er sindsdien wel 200.000 extra aandelen bij !!
    Al kan het best dat ze die bijkochten tussen nu en de lage $27

(NASDAQ: ARWR) today announced that it has filed an application for clearance to initiate a Phase 1/2a clinical trial of ARO-MMP7, the company’s investigational RNA interference (RNAi) therapeutic designed to reduce expression of matrix metalloproteinase 7 (MMP7) as a potential treatment for idiopathic pulmonary fibrosis (IPF).

    James Hamilton, M.D., MBA, senior vice president of discovery and translational medicine at Arrowhead, said: “Arrowhead’s inhaled pulmonary TRiMTM platform continues to generate compelling results in preclinical studies and promising drug candidates aimed at treating various diseases. ARO-MMP7 is a further expansion of our robust clinical pipeline, which also includes additional lung disease product candidates, ARO-RAGE and ARO-MUC5AC. MMP7 levels are a prognostic indicator in IPF, which is a progressive fibrotic disease characterized by a decline in lung function over time, and ARO-MMP7 offers a novel approach to potentially address the substantial unmet need in IPF.”

    An application for approval of the clinical trial was submitted to a local Ethics Committee and to the New Zealand Medicines and Medical Devices Safety Authority for review by the Standing Committee on Therapeutic Trials. Pending clearance, Arrowhead intends to proceed with AROMMP7-1001, a Phase 1/2a study to evaluate the safety, tolerability, pharmacokinetics, and pharmacodynamics of ARO-MMP7 in up to 56 healthy volunteers and in up to 21 patients with IPF.

    Weer een longindicatie erbij. Arrowhead moet zich wel erg zelfverzekerd voelen. Of is het meer een zaak van: vele malen schieten, dan is er vast wel eentje raak? We zullen het maar op het eerste houden. ;-)

    Maar waarom in Nieuw-Zeeland?

    [...] The biggest remaining scientific challenge is delivering Crispr to other organs, beyond the relatively easier areas of the blood, liver and eyes. Kiran Musunuru, a gene editing specialist at the University of Pennsylvania, said researchers are already starting to make some progress.

    “It’s going to blow up in the next 10 years,” he said. “That’s going to be transformative because it is going to greatly expand the number of diseases that we can treat with Crispr in patients.” 

    [...] Even more worrying to some investors were the trials paused because of safety concerns. Last year, the FDA halted several trials because of fears that the therapies could increase the risk of cancer. The most high profile were two by BlueBird Bio. One trial was restarted after the case of leukaemia in a participant was found to be unrelated to the drug. But in another, a link was made, though the regulator’s advisers have concluded the drug’s benefits still outweigh the risks, and it is waiting for approval to restart.

    Crispr companies stress that their technology is significantly different from conventional gene therapies, which involve delivering, rather than editing, a gene. John Evans, chief executive of Beam Therapeutics, said in some kinds of gene therapy you cannot always control where the gene is added, but with Crispr you can — and base editing, Beam’s specialism, is even more precise.

    Ik begin hem toch steeds meer te knijpen. Het is duidelijk dat velen hun pijlen op Crisper en consorten hebben gericht en daar gaat volgens mij het meeste geld naartoe, waardoor de ontwikkeling razendsnel gaat en het momentum meer en meer hun kant op valt. Rnai is prachtig, maar zonder meerdere, grote veiligheidsissues bij de Crispers gaat deze technologie het m.i. afleggen. De FDA staat al steeds meer toe en het lukt tegenstanders niet de Crispers aan banden te leggen. Dit is enkel tegen te houden door sae's. Zorgwekkend voor ons.

    Bij twijfel kan je ook toch in die technologie stappen ?

    Ik vertrouw verder op ARWR.
    De week die komt koop ik opnieuw een 125 aandelen extra bij, vermoedelijk midden tot einde deze week. afhankelijk wanneer de cash effectief op DeGiro staat.
    Zoals gewoonlijk iets te duur maar ik vertrouw op het komende najaar.
    Nog 4 maand straks en het jaar is alweer om.

    Natuurlijk kan er van alles roet in het eten komen gooien.
    Voldoende zaken : recessie , oorlog, energie,...

    Bioboyscout maakt zich de laatste tijd ook nogal "boos" op Twitter.

    Tuurlijk kan ik ook in BEAM of Intellia stappen, maar de waarderingen vind ik daar nog altijd te hoog.
    Ik vertrouw ook op ARWR, maar ik zie te veel 'krachten' naar m'n zin die willen dat gentherapie slaagt. Daar stroomt veel geld naartoe. Wetenschap is een beetje zoals de Formule 1. Waar het meeste geld zit, wordt het meeste succes geoogst...
