    momenteel zit ik klein in vrpx. Gister al fors gestegen (fda goedkeuring) maar stijgt aardig door.
    verder wil ik graag in OPGEN stappen. ook deze wacht op een fda goedkeuring. eind augustus.
    Zijn er mensen bekend met CLOVE HEALTH hier? dit aandeel probeer ik rustig bij te kopen. verwacht hier veel van in de toekomst.

(Nasdaq: SAVA), a biotechnology company focused on Alzheimer’s disease, announced today that it has reached agreement with the U.S. Food and Drug Administration (FDA) under a Special Protocol Assessment (SPA) for both of its pivotal Phase 3 studies of oral simufilam for the treatment of patients with Alzheimer’s disease.

    These SPA agreements document that FDA has reviewed and agreed upon the key design features of Cassava Sciences’ Phase 3 study protocols of simufilam for the treatment of patients with Alzheimer’s disease.

    “I believe these SPAs mark a meaningful and encouraging milestone for Cassava Sciences,” said Remi Barbier, President & CEO. “The SPAs underscore our alignment with FDA on key scientific, clinical and regulatory requirements of our Phase 3 program of simufilam in Alzheimer’s disease.”

    Cassava Sciences also reaffirmed prior guidance to advance simufilam into a Phase 3 pivotal program in Alzheimer’s disease in Fall 2021.

    The first clinical study protocol under the SPA is titled “A Phase 3, Randomized, Double-Blind, Placebo-Controlled, Parallel-Group, 52-Week Study Evaluating the Safety and Efficacy of One Dose of Simufilam in Subjects with Mild-to-Moderate Alzheimer's Disease”.

    The second clinical study protocol under the SPA is titled “A Phase 3, Randomized, Double-Blind, Placebo-Controlled, Parallel-Group, 76-Week Study Evaluating the Safety and Efficacy of Two Doses of Simufilam in Subjects with Mild-to-Moderate Alzheimer’s Disease”.

    Niet leuk natuurlijk zulke aantijgingen, maar we moeten dit niet groter maken. Voor nu totaal nog niets aan de hand. De tijd zal het leren, maar het shortersgilde haalt werkelijk alles uit de kast.

    Fiction: Biomarker data is generated by Cassava Sciences or its science collaborators and therefore are falsified.
    Fact: Cassava Sciences’ plasma p-tau data from Alzheimer’s patients was generated by Quanterix Corp., an independent company, and presented at the recent Alzheimer’s Association International Conference1.

    Fiction: Plasma p-tau for one individual Alzheimer’s patient increased by 235%, which was not shown in the scatterplot.
    Fact: This patient’s plasma p-tau increased by 38%, not 235%, as shown in a scatterplot.2

    Fiction: Tissue staining showing Abeta42 inside neurons shows treatment effects.
    Fact: Yes, Abeta42 is indeed inside neurons prior to plaque formation.

    Fiction: The author’s Citizen Petition to FDA dated August 18, 2021, is evidence of wrongdoing.
    Fact: Five days after the Citizen’s Petition, Cassava Sciences announced it had reached an agreement with FDA on Special Protocol Assessments (SPA) for its Phase 3 studies of simufilam for the treatment of Alzheimer’s disease.     Fact: Western blot analysis is foundational to the biotechnology industry5. Western blotting is a standard lab technique used world-wide to detect a protein of interest.

    Fiction: Cassava Sciences’ Western blots data appear overexposed and highly processed, evidence of image manipulation.
    Fact: High quality bands are supposed to look sharp6. Smudged bands can be evidence of inexperience, depending on levels of protein in the band.

    Fiction: Western blots data are identical, more evidence of image manipulation.
    Fact: The Western blots bands shown in the allegation are control bands. Control bands are supposed to be highly similar (since they show equal amounts of protein between lanes). Bands show clear differences when expanded. In addition, image manipulation of control bands makes no sense since these would not change the end data.

    Fiction: “Halo” effects in certain bands indicate fraud.
    Fact: A “Halo” effects in certain bands is a direct result of very dense dark loading control bands.7

    Fiction: Unusual looking bands on Western blots were pieced together from multiple sources.
    Fact: Proteins can and do stick to the side of a lane and migrate that way, resulting in ‘candy-wrapper’ appearance or other fictional images.

    Fiction: Femtomolar binding affinity is unusual and suspicious.
    Fact: Femtomolar binding affinity is a fundamental property of simufilam and may account for its relative potency and safety.

    Fiction: Post-mortem brain tissue that is dead for a decade is unreliable.
    Fact: Because of the inaccessibility of the human brain and its unavailability for biopsy, translational medicine can rely on post-mortem tissue8. In our case, human brain tissue was collected within 6 hours of death, flash-frozen and stored at -80 Centigrade. This is a standard procedure for pathologists. Such tissue processing is also used in cancer and other fields. Cassava Sciences is not aware of an industry-wide ‘expiration date’ on human post-mortem brain tissue that is properly collected, processed and stored.

    Fiction: Isoelectric focusing gels should not have crisp bands, which is evidence of fraud.
    Fact: Quality isoelectric focusing gels often do have crisp bands9.

    Fiction: Changes in the Y-maze test for transgenic mice could be interpreted as a decline in cognition.
    Fact: A panel of independent, peer-reviewers believe these changes represent an improvement, along with significant improvements in two other behavior tests.

    Fiction: High-affinity binding of naloxone for filamin A is suspicious.
    Fact: Naloxone binds the same site on filamin A. Of course, it will have high-affinity binding.

    Fiction: Isoelectric focusing experiments indicate 100% of filamin A is in altered conformation in Alzheimer’s disease and is largely restored to correct conformation by simufilam.
    Fact: Cassava Sciences agrees. This nicely describes the mechanism of action for simufilam.

    Haha, dat laatste is een mooie. Halen ze hun eigen nietszeggende claim onderuit :)

    Fictie: Iso-elektrische focusexperimenten geven aan dat 100% van filamine A zich in een veranderde conformatie bevindt bij de ziekte van Alzheimer en grotendeels wordt hersteld om de conformatie te corrigeren door simufilam.
    Feit: Cassave Sciences is het daarmee eens. Dit beschrijft mooi het werkingsmechanisme van simufilam.

    Goeie reactie van Cassava. De meeste aantijgingen snijden geen hout. De koers zal op korte termijn wel wat schade oplopen. Blijft over: een aantijging die ook bij mij toen al een beetje jeuk gaf: De fase 2b laboratoriumresultaten, die in eerste instantie niet interpreteerbaar waren, zijn later opnieuw geanalyseerd door een blijkbaar betrouwbaarder lab. Dat is een aanname.

    oppervlakkig gezien leest het meer als 'één grote nietes'

    Ps: zou mooi zijn voor toekomstige patienten als het allemaal meevalt, en ook voor de investeerders :)
    Maar de beschuldigingen zijn niet mals: datamanipulatie is voor mij eigenlijk toch wel een waar rampscenario bij elke biotech....zo verleidelijk maar zo fnuikend

    Weten we al meer over de achtergrond van deze klokkenluider?

    Ik heb zelf geen positie meer in Cassava, maar dat komt onder meer omdat ik gisteren afscheid heb genomen van mijn complete portefeuille. Het rendement vanaf 01-01-2021 was overall zo enorm hoog, dat ik er kriebels van kreeg. Al meermaals heb ik dat de afgelopen maanden zien verdampen. De afspraak die ik maakte was: bij een totale portefeuille van EUR XX,- verkoop ik alles en blijf ik aan de zijlijn staan. Mijn volledige portefeuille heb ik omgezet naar een schaduwportefeuille, zodat ik het verloop goed kan volgen.

    Of ik er goed aan heb gedaan? De tijd zal het leren.

    Helemaal met je eens, maar een enorm deel van mijn portefeuille heeft de afgelopen maanden/jaren echt gigantische rendementen opgeleverd. Amazon, Apple, Nvidia, ASMI, Google, Microsoft... het kon niet op.

    'De centen' kosten me op dit moment 0,7% per jaar, dus dat is minder. Ik hoop er gewoon op dat ik de pareltjes kan terugkopen tegen 10 tot 15% korting.

    Jij vindt dat je er aan goed gedaan hebt, en dan is dat zo ook. Iedereen heeft zijn eigen strategie, en ik snap heel goed wat je gedaan hebt. Ik heb dat met ASMI, Apple, Alfen en vooral ArgenX. Heb ook een hoop verkocht en een aantal saaie dividend aandelen voor gekocht, doe ik toch nog een beetje mee..;-)

    Meme Stock Cassava Plunges After Alzheimer’s Data Questioned

    (Bloomberg) -- Cassava Sciences Inc. plummeted 30% on Wednesday after a lawyer sought to stop studies of the biotech company’s experimental treatment for Alzheimer’s disease.

    A citizen petition from a former Securities and Exchange Commission enforcement lawyer questioned the quality and integrity of the results from the over 20-year-old company with no products on the market.

    “As a science company, we champion facts that can be evaluated and verified,” Remi Barbier, Cassava’s chief executive officer and founder, said in a statement countering the claims in the request.

    The stock had rallied more than 1,080% this year, gaining momentum on day trader enthusiasm for the space following the approval of Biogen Inc.’s Aduhelm, the first new drug approved for the brain disease in over a decade as well as Cassava’s own trial results.

    Lawyer Jordan Thomas of Labaton Sucharow alleged that some of the company’s results appeared to show signs of data manipulation, which Cassava denies.

    In response to the allegations Mayank Mamtani, an analyst with B. Riley Securities, recommended investors buy Cassava shares on weakness, seeing as the U.S. Food and Drug Administration has already reviewed the company’s data.“To our knowledge, citizen petitions are commonly associated with public endorsement (or lack thereof) of drug effects from experts and medical associations, rather than from law firms representing anonymous clients without disclosing conflict of interest,” he wrote.

    In February, Cassava said its experimental tablet, called simufilam, showed improvement in reasoning and behavior in a trial with less than 100 patients and without a control arm to gauge the impact of a placebo. That came after a data snafu whipsawed the stock last year when Cassava first reported a mid-stage study of simufilam had failed, but was later re-analyzed by a different lab which showed improvements in some biomarkers.

    On Tuesday, the company reached an agreement with U.S. regulators on the trial design for a much larger study which is expected to kick off this year.

    The petition could have some valid points, said Soumit Roy, an analyst with JonesTrading who has the highest price target on Wall Street at $215. But it is also potentially “over-interpreted with excessive digital analysis of figures published 15+ years ago,” he writes in a note to clients.

    “Several of the scientific articles brought into questions were published in reputed journals with rigorous peer review process. We would direct readers to Cassava to get finer details around each published figure,” Roy said.

    Bearish bets against the company have risen from a February low and now stand at $576 million, or roughly 13% of the company’s available shares, according to S3 Partners data.

    Austin, Texas-based Cassava didn’t respond to requests for comment.
