    Lyon, France; Leiden, the Netherlands, September 19, 2006 - Sanofi pasteur, the vaccines business of the sanofi-aventis Group, has within the framework of FLUPAN, a collaborative research project funded by the European Commission, generated the first clinical trial lot of a new generation of H7N1 pandemic vaccine. This trial will broaden sanofi pasteur's pandemic preparedness program initiated with the development of H5N1 vaccines.

    The H7N1 vaccine was produced at sanofi pasteur's Marcy l'Etoile facility in France using Crucell's PER.C6® cell-based technology, allowing an alternative production process expected to offer advantages over traditional manufacturing methods.

    This phase I clinical trial, initiated today in Bergen, Norway, is the first to assess the safety and ability to generate an immune response of a split, inactivated prototype pandemic H7N1 vaccine produced on cells.

    "The H7N1 vaccine strain was developed from an avian influenza virus (bird flu) by the UK's National Institute for Biological Standards and Control (NIBSC) and the University of Reading (UK). A highly pathogenic H7N1 avian influenza virus which caused outbreaks in Italian poultry in 1999 has been modified, so that it is safe to use and grows well in mammalian cell culture," commented Dr John Wood, FLUPAN Coordinator.

    Two influenza subtypes, H5 and H7, have recently caused highly pathogenic avian influenza. While most vaccine development has focused on H5N1 strains, avian H7-containing viruses remain a significant pandemic threat and have caused infections in humans in Europe over the last three years [1].

    "The FLUPAN coordinated study started today at the Haukeland University Hospital, University of Bergen (Norway). A total of 60 healthy adults aged 20 to 40 will be vaccinated with one of four formulations: two dosages (12 µg and 24 µg) with or without an aluminum hydroxide adjuvant, an additive commonly used to increase the immune response to vaccines," said Dr Lars R. Haaheim, Principal Clinical Investigator, University of Bergen.

    Sanofi pasteur selected PER.C6® cells for their high susceptibility to influenza viruses, thereby making the production of large amounts of influenza vaccine feasible for both pandemic and seasonal strains.

    Pandemic Influenza Overview
    Influenza is a highly infectious virus that spreads easily from person to person, primarily when an infected individual coughs or sneezes. An influenza pandemic is a global epidemic of an especially virulent virus, newly infectious for humans, with the potential to cause severe morbidity and mortality. According to the World Health Organization (WHO), the next pandemic is likely to result in 1 to 2.3 million hospitalizations and 280,000 to 650,000 deaths in industrialized nations alone. Its impact is expected to be even more devastating in developing countries. In an attempt to minimize the impact of a pandemic, many countries are developing national and transnational plans against an eventual influenza pandemic situation.

    Several strains of influenza virus are potentially pandemic. An H7N7 strain was responsible for a large outbreak of fowl plague in the Netherlands in 2003 that spread to 89 humans. While symptoms were mostly mild, one case died from pneumonia in combination with acute respiratory distress syndrome as a result of the infection. The virus was also seen to be transmitted to family contacts[1, 2].

    FLUPAN news – September 2006

    The project began on 1 September 2001 and the scientific progress can be summarised as follows:

    A highly pathogenic H7N1 avian influenza virus was selected as a potential pandemic virus. This virus caused lethal outbreaks in Italian poultry in 1999 and was subsequently shown to be related to the 2003 H7N7 poultry virus in the Netherlands, which was responsible for over 80 human infections and one death.
    Using reverse genetics, The H7 haemagglutinin (HA) of the Italian H7N1 virus was modified so that the virus would be non pathogenic.
    A reassortant virus bearing the modified H7 HA and the N1 neuraminidase (RD-3) has been prepared.
    The RD-3 virus has passed internationally-agreed safety tests has been used to produce an inactivated vaccine on the PER.C6 mammalian cell line.
    A clinical trial of the H7N1 vaccine will begin in September 2006.
    Surveillance of wild and domestic birds in Italy has shown that influenza viruses from several different subtypes (including non-pathogenic H5 and H7) have been in circulation.
    A modified serological test has been developed with increased sensitivity for detection of antibody to H5 and H7 influenza viruses. This test can be used in serosurveillance to provide early warning of a possible pandemic. It also will be useful in vaccine evaluation.
    Avian influenza viruses from surveillance in Italy have provided a basis for preparation of a library of reagents, which will be a valuable resource for pandemic vaccine development in the future.

    What are the objectives of FLUPAN?

    There are six European partners in FLUPAN: National Institute for Biological Standards and Control, UK; University of Reading, UK; Istituto Superiore di Sanita, Italy; Heath Protection Agency, UK; sanofi pasteur, France; University of Bergen, Norway. Reverse genetics will be used to modify a highly pathogenic H7N1 avian influenza virus, so that it is safe to use and will grow well on mammalian cells. Experimental vaccines will be produced, which will be evaluated pre-clinically and clinically. Libraries of reagents will be produced from a variety of animal influenza viruses and new techniques will be investigated for producing cell culture vaccines and for monitoring transmission of animal viruses to man. The project should improve the level of pandemic preparation in the European Community.

    Previously, HHS awarded Sanofi Pasteur a $97 million contract for development of a cell-based vaccine in April 2005. This company was the first to be awarded a federal contract for commercial scale production of newer influenza vaccine methodology.

    Met een dikke financiële injectie zal dat wel meevallen geel.

    Er is nog maar 2,3 miljard te verkrijgen. Jammer voorde andere partijen!

    OTee: Klopt, daarom ook koos men voor de H7N7 strain ...
    Volgens de bijbel worden cycli van 7 magere jaren afgewisseld met cycli van 7 vette jaren, maar volgens sceptici is het allemaal onzin. Alleen Flip-Flap's hechten er geloof aan.

    Hi Giraf,

    Duidelijk is dat de belegger die op zekerheid koerst, op bevesting zit te wachten. Voor de insider zijn deze PBs geen verrassing (want door het management aangegeven dat ze eraan komen in deze periode).

    Wij weten dat Q en de US miljoenen eraan zitten te komen! Het PB dat dit bekend gaan maken, zal de koers flink opjagen.

    Heren en dames koersmakers.

    Willen jullie dit stukje tekst nogmaals goed door lezen? Tot 10x toe? Wellicht dat jullie dan in de gaten hebben wat voor goudklompje er voor jullie klaar ligt.

    Mochten jullie het desalniettemin niet begrijpen.... Biocon, Biostefan, Joie en vele anderen op dit forum (zoals ik zei desnoods de gek) zijn van harte bereid om het e.e.a. uit te leggen over deze lastige materie die Bio-tech heet. Of bel anders even met Marcel Wijma. Die snapt de materie tenminste wel. :-)

    Pracht van een bericht..... zou minstens een koerssprong van 1 euro moeten veroorzaken. Voor hen die het op waarde weten te schatten.

    Inderdaad baanbrekend nieuws Jahn1, velen schijnen niet te beseffen dat we hier spreken over dé Sanofi-Aventis, diezelfde die nu al jaarlijks goed is voor ongeveer de helft van de te leveren griepvaccins aan de VS (mégamarkt), en was diegene die veruit de andere helft levert niet Chiron ?, nu overgenomen door Novartis Ag, precies ja,.. diegene die vorige week nog een Star-deal afsloot met "onze" Crucell ! (snap je 50% + 50%)....DDS-scenario zet zich om in langspeelfim

    Hoi Jan,

    Ik vrees dat de genoemde groep weer eens even langs de zijlijn blijft staan. Was in het verleden een samenwerking met Sanofi al voldoende om de koers op een dag dusdanig te laten stijgen dat er een "freeze" moest worden ingebouwd, nu laten ze het afweten. We raken verwend hier met ons allen. Geen nieuws is goed genoeg meer.

    Terwijl dit persbericht zoveel potentie heeft. Het is niet alleen dat het e.e.a. onderzocht wordt. In tegendeel..... uit onderzoek blijkt nu dat het werkt. Belangrijk gegeven...

    Ben ik nu de enige die het snapt?
