    ASADENA, Calif.--(BUSINESS WIRE)--Apr. 15, 2021-- Arrowhead Pharmaceuticals, Inc. (NASDAQ: ARWR) today announced ARO-DUX4 as Arrowhead’s first muscle targeted investigational RNAi therapeutic candidate to utilize its proprietary Targeted RNAi Molecule (TRIMTM) platform. ARO-DUX4 is designed to target the gene that encodes human double homeobox 4 (DUX4) protein as a potential treatment for patients with facioscapulohumeral muscular dystrophy (FSHD). Pending abstract acceptance, Arrowhead intends to present preclinical data on ARO-DUX4 at the 28th Annual FSHD Society International Research Congress being held virtually on June 24-25, 2021. Arrowhead is currently conducting IND/CTA enabling toxicology studies and intends to file for regulatory clearance in the third quarter of 2021 to begin clinical studies of ARO-DUX4.

    Chris Anzalone, Ph.D., president and chief executive officer at Arrowhead, said: “ARO-DUX4 represents further expansion of our pipeline into diseases of the skeletal muscle, which is the fourth cell type that we may potentially be able to target with our proprietary TRiMTM platform. In addition, DUX4 fits perfectly with our strategy not only to bring RNAi outside the liver, but also to select gene targets that we believe are clear causes of specific diseases and for which there is strong biologic and genetic validation. We believe our preclinical data, which we intend to present at the FSHD Society International Research Congress in June pending abstract acceptance, highly support progressing ARO-DUX4 into clinical studies. In our various animal models, ARO-DUX4 reduced DUX4 expression by greater than 70%, prevented body weight loss associated with tamoxifen-induced DUX4 expression, and prevented loss of muscle function.”

    Great point and very astute analysis! IMO, if they didn't have proof-of-concept for tumor and lung, I highly doubt they'd be announcing muscle right now, as it would probably be placed on hold.

    Goed punt van BioBoyScout. Dank Wil om dat onder de aandacht te brengen. Het zou zeker een reden kunnen zijn, maar zelf dacht ik meer in strategische richting. CA zal niet meteen met het grote nieuws willen starten. Eerst de markt polsen met iets relatief kleins, en pas als de omstandigheden ideaal zijn de kanonnen laten knallen. :-)

    A small molecule for alpha-1 antitrypsin deficiency faces a key clinical test, and its chances of success aren’t good.

    Zou leuk zijn als dit idd mislukt. Geeft nog meer opwaartse stuwing aan de koers van Arrowhead.

    Ik zat er gelukkig naast. Dit is wat CA in Q2, 2020 te melden had:

    "If the data are supportive of further development we hope to launch a
    Phase 3 study in 2021 and move to rapidly expand the pipeline with product
    candidates that address other underserved pulmonary diseases, such as COPD,
    asthma, and pulmonary fibrosis.
    Toward that goal, I’m pleased to announce that we have completed discovery and
    optimization work on our second lung-targeted program and have nominated
    ARO-Lung2 as our next candidate. We are not disclosing the target at this point,
    but we can say that it is designed to address COPD patients. We have been very
    encouraged by the data in this program and for ARO-ENaC and are eager to begin
    the expansion of our pulmonary franchise. We are now working on manufacturing
    and IND-enabling toxicology studies of ARO-Lung2 and plan on filing a CTA in
    the first half of 2021 to begin first-in-human studies. We had previously hoped
    this could happen by the end of the year, but COVID has slowed development
    down a bit and will delay first in human studies by probably 1 or 2 quarters."
