Veklury should only be administered in a hospital or in a healthcare setting capable of providing acute care comparable to inpatient hospital care. Veklury is the first treatment for COVID-19 to receive FDA approval.

House, SA News Editor The FDA's full approval of Gilead Sciences' (NASDAQ:GILD) Veklury (remdesivir) on Thursday, October 22, for severely ill hospitalized COVID-19 patients was the first full nod for such a therapy and sustains the company's leadership position in antiviral treatments against SARS-CoV-2 infection. Remdesivir was also the first antiviral treatment to receive emergency use authorization (EUA) in the U.S., Europe, Canada, Australia and Japan to treat the respiratory infection. The company is actively working to expand the RNA polymerase inhibitor's indications. Its current U.S. EUA covers moderately ill patients and hospitalized pediatric patients under the age of 12 with certain weight restrictions. A search in ClinicalTrials.gov listed 23 ongoing or planned clinical trials on remdesivir. Four studies involve pediatric patients including a company-sponsored 52-subject Phase 2/3 study, CARAVAN, evaluating the drug in young people less than 18 years old. Primary endpoints are safety and pharmacokinetics. Secondary endpoints include efficacy measures. The estimated completion date is February 2021. Gilead is also sponsoring a 1,230-subject Phase 3 trial assessing remdesivir's effect on reducing the rate of hospitalization or death in outpatients as young as 12 years old with COVID-19. The primary endpoints are safety and the proportion of hospitalizations or death by day 14. The primary completion date is December. Yet another company-sponsored trial is a 282-subject Phase 1/2 evaluating an inhalable formulation of the antiviral in adult healthy volunteers (first part) then adult patients with early-stage infection. The primary endpoint of the second part is the time-weighted average change from baseline in SARS-CoV-2 viral load at day 7. The estimated completion date is also December. Another key study, sponsored by Roche (OTCQX:RHHBY), is a 450-subject Phase 3 trial, REMDACTA, assessing remdesivir plus Actemra (tocilizumab) compared to placebo plus tocilizumab in hospitalized patients as young as 12 years old with severe COVID-19. The primary endpoint is clinical status at day 28. The estimated completion date is December 1. NIH's National Institute of Allergy and Infectious Diseases (NIAID) is conducting two adaptive late-stage studies, ACTT-2, assessing the combination of remdesivir and Eli Lilly's (NYSE:LLY) Olumiant (baricitinib) compared to remdesivir alone in adult hospitalized patients and ACTT-3, evaluating remdesivir plus interferon beta-1a compared to remdesivir alone. The estimated completion dates are August 2023 and November 2023, respectively. Als een ernstige infusiegerelateerde overgevoeligheidsreactie optreedt, stop dan onmiddellijk met Veklury en start een geschikte behandeling (zie rubriek 4.3). Verhoogd risico op transaminaseverhogingen: Transaminaseverhogingen zijn waargenomen bij gezonde vrijwilligers en bij patiënten met COVID-19 die Veklury kregen; deze verhogingen zijn ook gemeld als een klinisch kenmerk van COVID-19. Voer bij alle patiënten hepatische laboratoriumtesten uit (zie Dosering en toediening). Overweeg om met Veklury te stoppen als de ALAT-waarden stijgen tot> 10x ULN. Stop met Veklury als ALAT-verhoging gepaard gaat met tekenen of symptomen van leverontsteking. Risico op verminderde antivirale activiteit bij gelijktijdige toediening met chloroquine of hydroxychloroquine: Gelijktijdige toediening van Veklury met chloroquine-fosfaat of hydroxychloroquinesulfaat wordt niet aanbevolen vanwege antagonisme waargenomen in celkweek, wat kan leiden tot een afname van de antivirale activiteit van Veklury. Remdesivir is owned by Gilead Sciences (NASDAQ:GILD) and the lipid nanocrystal (LNC) platform is owned by Matinas. Any product generated as a part of the partnership would require a license from Gilead and Matinas for the use of remdesivir and LNC formulation, respectively. One or more formulations of remdesivir will be developed using Matinas’ LNC platform delivery technology, which enables the development of a wide range of difficult-to-deliver molecules. Matinas plans to utilize NIAID’s suite of preclinical services to carry out antiviral testing. (GILD)By: Mamta Mayani, SA News Editor4 Comments Kite, a Gilead Company (NASDAQ:GILD), announces four-year follow-up data from the pivotal ZUMA-1 trial of Yescarta (axicabtagene ciloleucel) in adult patients with refractory large B-cell lymphoma (LBCL). Among Yescarta-treated patients (n=101) with a minimum follow-up of four years (median follow-up of 51.1 months), the Kaplan-Meier estimate of the four-year overall survival (OS) rate was 44%. The median time from leukapheresis to complete response (CR) was less than two months. No Yescarta-related secondary malignancies were reported. Blood sample analyses provided by 21 patients, showing an ongoing response at three years follow-up also demonstrated that 67% (n=14/21) had detectable CAR gene-marked cells and polyclonal B cells in blood. Normal B-cells were present in 91% (n=21/23) of evaluable patients. These results suggest that persistence of functional CAR T cells is not necessary for durable remissions in patients with refractory LBCL and may support long-term safety of the therapy. The data were presented at the 62nd American Society of Hematology (ASH) Annual Meeting and Exposition.

April 26, 2021 Gilead Sciences Announces Steps to Expand Availability of Remdesivir in India – Gilead Will Donate a Minimum of 450,000 Vials of Veklury® to the Government of India – – Company Will Continue to Support Expansion of Local Production Capacity – FOSTER CITY, Calif.--(BUSINESS WIRE)-- Gilead Sciences, Inc. (Nasdaq: GILD) today announced that in response to the rapid increase in COVID-19 cases in India, the company is providing its voluntary licensing partners with technical assistance, support for the addition of new local manufacturing facilities and the donation of active pharmaceutical ingredient (API) to rapidly scale up production of remdesivir. Remdesivir is approved in India for restricted emergency use for the treatment of suspected or laboratory confirmed COVID-19 in adults and children hospitalized with severe disease. In addition to providing support to its licensees to expand their local manufacturing capacity, Gilead will also donate at least 450,000 vials of Veklury® (remdesivir) to help address the immediate needs of Indian patients. “The recent surge of COVID-19 cases in India is having a devastating impact on communities and has created unprecedented pressure on health systems,” said Johanna Mercier, Chief Commercial Officer, Gilead Sciences. “We are committed to doing our part to help tackle this crisis.
