ABM Financial News - 09/07/2018 (ABM FN-Dow Jones) Stifel heeft maandag het koersdoel voor Pharming Group verhoogd van 1,55 naar 1,80 euro met een ongewijzigde koopaanbeveling. De analisten besloten de taxaties voor de toepassing voor de ziekte van Pompe toe te voegen in model. In aanloop naar de cijfers over het tweede kwartaal op 26 juli verhoogde Stifel daarnaast de taxaties ook vanwege de verkoop van Ruconest. Hoewel de omzet afgelopen drie maanden vermoedelijk gelijk was aan die in het eerste kwartaal, betekent dit op jaarbasis volgens de marktvorsers nog altijd een groei van 93 procent. Momenteel vindt Stifel dat het een goed moment is om aandelen Pharming te kopen. De analisten mikken op een positieve nieuwsstroom, terwijl het biotechbedrijf de strategie voor Ruconest uitrolt en het eigen platform aan het werk zet. Wainwright heeft vrijdag het koersdoel voor Pharming Group verhoogd van 2,10 naar 3,10 euro met een onveranderde koopaanbeveling. De koersdoelverhoging was een reactie op de donderdag gehouden beleggersdag door Pharming in New York. "Wij waren onder de indruk van de visibiliteit die de onderneming verstrekte", aldus analist Joseph Pantginias. Vooral de nieuwe initiatieven die Pharming presenteerde konden op goedkeuring van de analist rekenen. "Er worden meer zuigers aan de motor toegevoegd", oordeelde Pantginias. Pharming legde in New York onder meer uit waarom een middel als Ruconest gunstige therapeutische effecten kan hebben op pre-eclampsie, zwangerschapsvergiftiging, volgens de nieuwste inzichten over de ontwikkeling van deze aandoening. Op het vlak van contrast-geinduceerde nephropatie, een nieraandoening, is Pharming al een fase II-studie aan het doen in het Zwitserse Basel. Ook gaf Pharming een update over de ontwikkeling van proteïnevervangende producten voor de ziekte van Pompe en de ziekte van Fabry. Deze producten zouden aanzienlijke beperkingen van bestaande behandelingsmethodes aanpakken. Het aandeel Pharming handelde vrijdag 0,5 procent hoger op 1,51 euro. Van de hele geplande grote investors tour waarmee Pharming de 17e start, hoef ik eigenlijk maar een ding te weten. Wanneer kunnen Pharming's HAE patiënten eindelijk van het infuus af om over te kunnen stappen naar de nieuwe vorm van een injectie of een plijster? Het tijdsplaatje is hier het enige van belang. Voor de rest verwacht ik een herhaling van dezelfde ouwe meuk uit Ney York. Maar dat is natuurlijk niet te hopen, en niet waarop zovelen zitten te wachten.

Is één van de KT zaken die Ph moet binnenhalen evenals EMA (<12 jr), Basel (uitbreiding toepassing) en FDA (prfylaxe) om het gebrek aan significante winststijging te compenseren. Anders zie ik het met de koers de rest van dit jaar somber in. LT zaken (pijplijn) hebben geen enkel effect meer op de koers, die mooie tijd is na febr 2018 (ong.) voorbij.

Concurrentie van Shire's nieuwe product zou het marktaandeel van Pharming's Ruconest aantasten. Volgende week vrijdag 21 september hoort Pharming of het in de VS wel of niet Ruconest mag aanbieden voor preventief gebruik. CRL Delays Prophylaxis Approval; Long-Term Story Remains Intact SUMMARY Pharming (PHARM) announced the receipt of a Complete Response Letter (CRL) regarding its sBLA for Ruconest use in prophylaxis for hereditary angioedema (HAE, currently marketed in acute), ahead of the 9/21 PDUFA. The FDA requested a Phase 3 study be conducted before it will reevaluate Ruconest in prophylaxis; we expect a 12-24-month delay. While this outcome is surprising, we believe today's selloff (down ~20%) is overdone. There is little impact to medium-term sales in our model (prophylaxis had constituted only 3% of 2021E revenues), and we believe the longterm story remains intact. We push out prophylaxis sales and conservatively lower acute sales estimates as Shire's Takhzyro (lanadelumab) launch and Cinryze supply stabilization could slow Ruconest penetration. We lower our PT to €2.50 (from €3.00). KEY POINTS ¦ The sBLA was filed on the basis of two Phase 2 studies in prophylaxis (one open label and one randomized, double-blind, placebo-controlled, total n=56) and an end-of-Phase 2 meeting. Given the supportive feedback from the FDA, a highprofile Lancet publication and the strength of the Phase 2 data, both the company and the Street had expected an approval (80% probability). ¦ We await further clarity once PHARM has discussed the CRL and potential resolutions with the FDA. One scenario would be running the Phase 3 study with next-generation formulations of Ruconest (subQ, intradermal) already in development rather than taking the current IV formulation forward. We expect a Phase 3 study of that nature could initiate mid-/2H19 (12- or 26-week duration). ¦ With prophylaxis approval delayed, investors will be focused on the sales trajectory for Ruconest and quarterly execution in 2H18-19. We reduce our 2019E and onward prophylaxis numbers and expect very minor off-label usage ($1M to $5M per year). With a clean lanadelumab label and stabilized Cinryze production constraints on plasma-derived C1C1 esterase inhibitor, we reduce our 2019Eonward acute Ruconest sales estimates. ¦ PHARM's pipeline for Ruconest in indications outside of HAE is also increasingly important. The Phase 2 IST for cardiac-induced nephropathy (CIN) will report data in 4Q18 (go/no-go for company-sponsored Phase 2 in 1H19), and a Phase 1/2 study for pre-eclampsia could initiate as soon as YE18. Finally, a Phase 2 study in cardiac protection is planned for 2H19. ¦ We expect continued negativity and pressure on the shares as noise around competition, a strong Takhyzro label/launch, and the prophylaxis disappointment flush out. However, long-term investors should note the impressive Ruconest US sales growth in acute treatment, multitude of pipeline catalysts, and prudent OPEX spend (potentially break-even in 2018). We stay bullish.
STIFEL. BUY TARGET PRICE CHANGE September 20, 2018 EU Pharmaceuticals Pharming Group N.V. PHARM – AEX; PHAR.AS No prophylaxis approval but sell-off overdone Summary Pharming has received a complete response letter (CRL) from the FDA to expand the use of Ruconest for the prophylaxis treatment of HAE, beyond the existing use in the acute setting. We are surprised by the FDA's decision and request for a further clinical trial, given the relative strength of the clinical data compared to Cinryze as one of the approved drugs for prophylaxis use. We can only speculate that the more recent approvals for Haegarda and Takhzyro have moved the FDA's goalposts on what data are required. While approval for Ruconest in the prophylaxis setting was not a game-changer given the focus on Ruconest as the best option for acute, rational, on demand treatment, we did expect a boost to sales growth in 2019 from renewed sales force promotion and have therefore pushed back our sales contribution from prophylaxis use to 2021. This lowers our 2022 sales forecast from $425m to $350m, and therefore our TP is reduced to €1.45. We believe the recent sell-off has been overdone and therefore reiterate our Buy rating. Key Points Surprising FDA rejection, Ruconest clearly works and is safe. Pharming has previously reported, presented and published (in The Lancet) highly positive Phase II clinical data with the use of Ruconest for the prophylaxis treatment of HAE. A onceweekly and twice-weekly IV regimen resulted in significant reductions in HAE attacks vs placebo, with what we also regard was a greater response rate (>50% reduction in attacks) compared to Cinryze. The data demonstrate the product clearly works in the prophylaxis setting, has been on the market since 2014 as an acute treatment, has an established/acceptable safety profile, and anecdotally has been used as a prophylaxis option, particularly during the recent shortages of Cinryze. The FDA's decision to ask for another clinical trial, just to confirm a treatment response already observed, is bizarre in our view. Our only conclusion is that the FDA has taken a wider market view in light of approvals for Haegarda and Takhzyro as new prophylaxis treatments and sees less need to sanction an accelerated approval (i.e. on Phase II data). Options for next steps. We understand that Pharming needs to have a follow-up meeting with the FDA to understand what clinical data the regulator requires in order to seek prophylaxis approval. We see little merit in proceeding with the current IV regimen for the next clinical trial, instead waiting for more convenient formulations (SC, IM, transdermal patch), for a more appropriate offering in the prophylaxis setting. We therefore have pushed back potential sales contribution from use in the prophylaxis setting until 2021, which lowers our 2022 sales estimate to $350m from $425m. Even with shifting dynamics, significant acute opportunity remains. We do expect the overall HAE market to shift to more convenient prophylaxis products, such as Haegarda (twice-weekly, SC), Takhzyro (once every 2 weeks, SC) and potentially daily oral tablets (e.g. BioCryst's BCX7353 in Phase III), with a 40:60 proph:acute ratio today to a 75:25 split in favour of proph by 2022. However, that still leaves a significant market opportunity, we estimate at around $700m, and assuming Ruconest becomes available in more convenient formulations, we believe Ruconest will have the best profile in the acute setting, as a standard regimen but also to treat breakthrough attacks which will inevitably occur in patients on prophylactic options. A head-to-head trial of Ruconest vs Firazyr due by the end of 2018 will also be important given the likely genericisation of Firazyr by July 2019. Ultimately, we also expect the likes of Takyzyro to help to expand the market for HAE treatment, as Shire anticipates, rather than displacing existing treatments, i.e. more market expansion than market rotation. Sell-off overdone. At the core of Pharming is a validated and differentiated technology platform that has so far delivered Ruconest and has the potential to deliver further candidates, such as for Pompe disease, due to enter the clinic by year-end. In our view the recent sell-off presents an attractive buying opportunity given overblown concerns over the headwinds to Ruconest.

CRL received ahead of Friday's PDUFA. This morning, Pharming announced that it received a Complete Response Letter (CRL) from the FDA last night regarding Ruconest's sBLA for the prophylaxis setting. Recall that the PDUFA date was this Friday, September 21. The sBLA was accepted for review in January 2018 based on the additional positive randomized Phase 2 in the prophylaxis setting, supported by already approved indication in the acute setting for the drug and its sizable database of efficacy and safety. The FDA has requested an additional trial to "further evaluate the effectiveness of Ruconest in HAE patient". While not disclosed, we believe that the CRL could be requesting further clinical information on the breakout of subpopulations tested, based on site of attacks (e.g., GI, upper airway, subcutaneous/ skin), but this remains to be seen. So now what? Big help that Ruconest already on the market with growing sales. We believe that today's market reaction, especially in the E.U., is an overreaction to the receipt of the CRL. While discussed below, the main impact to the thesis is impacted only through the NPV of our prophylaxis projections, specifically projected launch timing. Further, we believe that the delineation between acute and prophylaxis is not completely black and white, as Ruconest is already on the market in the early phases of its strong growth in North America. To this end, we believe that the drug is being used by physicians and patients collaborate in such a fashion to use Ruconest in ways to best manage their individual patterns of HAE attacks. Regarding next steps, we discussed this with management this morning, and while they have internal undisclosed views on this, are going to take deliberate and temperate steps to discuss plans with the FDA. Since this work has also been ongoing in earnest, it appears to be an opportune time to include the potential for the improved formulations of Ruconest in the discussion with the FDA (i.e., subcutaneous and intramuscular, which could significantly improve patient convenience). So as of now, the company is in a bit of a regroup as it plans for discussions with the FDA to define the next clinical step, while it continues to deliver on new and more convenient formulations, as well as continuing to enjoy growing North American revenue and market traction for Ruconest. Formulation work important to improving thesis. Pharming’s efforts to increase the convenience and control of dosing for Ruconest in patients with HAE are ongoing. These efforts include: (1) line extension vial containing 1,000 U for pediatric use; (2) "Ruconest lite" vial of 2,100 U of lyophilized drug with shorter reconstitution times (3 min. vs. 5-6 min.); IND-enabling studies ongoing; and (3) "Ruconest Liquid" ready to use 3 ml vial of >500 U/ml; in final development for convenience factor. The 'lite' and 'liquid' versions could be used for IV use, but importantly IM for acute attacks or SC or ID for prophylaxis. An ID formulation could also be combined with a new proprietary painless ID delivery system, which includes a dissolving point device, and a reservoir device, which should differentiate against the competition, all of which have painful injections. Ultimately we believe the strategy to offer multiple options of administration with the goal of providing flexibility for the patients could represent a significant advantage for the company among competitors.

vandaag contact gehad met van Lanschot Kempen, zoals de toko volledig heet. Volgens Mifid 2, ken uw klant is daarin belangrijk, wil van L Kempen het rapport niet afgeven. Rapport was en is alleen bedoeld als grote instit. beleggers en is was niet voor 3e bested. Allemaal heel vreemd, nog vreemder is het dat van L Kempen op de beurs meer dan een euro stijgt. Hoewel vreemd, nee. Zij hebben een hoop geld verdiend als comm. op grote verkoop en later kooporders. Maar het is zeer beschamend dat een analist niet vooraf met een CEO heft gesproken. Aan dit soort analisten geleuter moet echt een eind aan komen. De een schrijft verkopen bijv Besi, een paar dagen later schrijft een andere analist kopen bijv Besi. dit is een heel mooi voorbeeld, kortom het lijkt het KNMI wel. Stuk in de Telegraaf vanochtend was een redelijk positief stuk, een stuk wat in ieder geval onderbouwd is en waarvan er overleg of terkennis giving aan de CEO is gevraagd/gegeven. Op 5 oktober is er een BAVA in Den Bosch van van L Kempen, zou mooi zijn als er een stel Brabo heen zou gaan en in de vergadering iedere keer het rapport Pharming zouden aanhalen. Oja in tegenstelling tot ING heft de CEO van van L Kempen wel een zeer forse salarisverhoging gevraagd en gekregen. Hallo, waar is Wopke??
