    Mooi dat zei o.a. ook hierover beginnen (eigenlijk mijn enige zorg bij Genmab): De explosieve groei van allemaal units (marketing etc) die niks met de kern van Genmab (ontwikkeling van 'knock your socks of' antilichamen) te maken hebben. Ik hoop natuurlijk dat het allemaal goed uitpakt maar na de koop van die fabriek in 2008 ben ik nog wel op mijn hoede :) Als er dit jaar een dikke cashburn is en de verkoopcijfers vallen enigzins tegen dan is dat toch een behoorlijk risico voor de koers denk ik. Ik zou graag willen dat ze dit nog wat beter argumenteren ik vind dit niet terug in het jaarverslag.

    Al een paar dagen online, wel positief nieuws en ook focus op effecten in solids.

    "Seattle Genetics, Inc. (Nasdaq: SGEN) today announced dosing of the first patient in the phase 2 innovaTV 204 clinical trial evaluating the efficacy, safety and tolerability of tisotumab vedotin as monotherapy for patients with recurrent and/or metastatic cervical cancer who have relapsed or progressed after standard of care treatment.

    Tisotumab vedotin is being developed in collaboration with Genmab A/S. The innovaTV 204 trial is intended to support potential registration under the U.S. Food and Drug Administration's (FDA) accelerated approval regulations. Tisotumab vedotin is an investigational antibody-drug conjugate (ADC) designed to target Tissue Factor antigen on cancer cell surfaces and deliver the cell-killing agent monomethyl auristatin E (MMAE) directly inside cancer cells. The Tissue Factor antigen target is overexpressed in the vast majority of patients with cervical cancer and in many other solid tumors, including ovarian, lung, pancreatic, colorectal and head and neck.

    'While significant advances have been made in the treatment and prevention of cervical cancer, there remains a need for targeted agents that are effective for patients who progress or relapse after standard treatments,' said Robert Coleman, M.D., FACOG, FACS, Professor of Gynecologic Oncology, The University of Texas MD Anderson Cancer Center, Houston, TXand principal investigator of the innovaTV 204 trial. 'In a prior study, tisotumab vedotin demonstrated encouraging results in previously treated recurrent and/or metastatic cervical cancer, an area of unmet need where there is no established standard of care and response rates are limited.'

    'With the initiation of this phase 2 study of tisotumab vedotin for women with previously treated recurrent and/or metastatic cervical cancer, we now have four late-stage development programs on registrational pathways to achieve our goal of becoming a global, multi-product oncology company,' said Roger Dansey, M.D., Chief Medical Officer at Seattle Genetics. 'In addition, we plan to evaluate tisotumab vedotin in collaboration with Genmab in other solid tumors where there remains an unmet medical need for new treatment options.'
    The phase 2 innovaTV 204 study (also known as GCT1015-04) is a global, multicenter, single arm trial that will enroll approximately 100 patients with recurrent and/or metastatic (2nd and 3rd line) cervical cancer who progressed on or relapsed after treatment with platinum-based chemotherapy used alone or in combination with bevacizumab (Avastin). Patients will be treated with single-agent tisotumab vedotin every three weeks. The primary endpoint of the trial is objective response rate as assessed by independent review. Key secondary endpoints include duration of response, progression-free survival, overall survival, safety and tolerability.

    The companies also plan to evaluate tisotumab vedotin in multiple solid tumors in a phase 2 trial this year.

    Core business productgroei zet mooi door, gezien cijfers mei. En wat betreft opzetten (marketing) units, dat is zowel een kans als een risico. Genmab wil met verdiensten Darzalex een meer onafhankelijk bedrijf opbouwen, met hoger percentage voor zichzelf en (deels) dit dan ook substance geven door eigen verkooporganisatie op te bouwen. Is direct, puur eigen sales, en indirect, in toekomstige onderhandelingen met big pharma over percentages bruikbaar. Wel moeten we hun focus in de gaten houden, want het zal moeilijk zijn de salespower van de big pharma's te matchen. Moet je ook niet willen. Als ze de organisatie inrichten om o.a. medicijnen te verkopen die (net) niet interessant genoeg zijn voor big pharma, maar wel voor Genmab, in kader van portfolio diversificatie, dan zit er wat in. Kortom, deze aanpak is een mixed bag.

    Is naar aanleiding van GS presentatie van vd Winkel onlangs. Bottom line: schrijf toepassing solid tumors zeker nog niet af. Diverse variant studies lopen nog door en men is druk de data van de gestopte studie aan het analyseren. Ach, beter het solid verhaal afschrijven en dat het later meevalt...

    Nieuwe milestone in collab met Seattle Genetics. Dit is dus de samenwerking waarbij Genmab, bij een succesvolle ontwikkeling van een nieuw medicijn, dat op de markt komt, het grootste deel van de koek behoudt. Betreft overigens trial in (7!) solid tumors.
    In dit geval betaalt overigens Genmab de milestonebetaling. Lees hele bericht hierna. Dit kan interessant worden, maar nog wel even te gaan aangezien nu fase I/II.

    Copenhagen, Denmark; June 29, 2018 – Genmab A/S (Nasdaq Copenhagen: GEN) announced today that it will pay a milestone payment of USD 7 million to Seattle Genetics, Inc. (Nasdaq: SGEN) following progress in the HuMax-AXL-ADC development program. The milestone payment was triggered by the initiation of expansion cohorts in the ongoing Phase I/II trial in solid tumors. Genmab originally licensed Seattle Genetics’ ADC technology in September 2014, in order to combine it with Genmab’s proprietary HuMax-AXL antibody to target multiple tumor types.

    “This milestone payment marks the rapid progress of the development program for HuMax-AXL-ADC and we very much look forward to seeing data from this program in the future,” said Jan van de Winkel, Ph.D., Chief Executive Officer of Genmab. “This is the second development interaction we have with Seattle Genetics, who are experts in the field of antibody-drug conjugates. The licensing deal for the use of their ADC technology in our proprietary HuMax-AXL-ADC program allows us to pursue our goal of developing truly differentiated cancer therapeutics whilst retaining maximal ownership of our therapeutic products.”

    Today's news does not impact Genmab's 2018 financial guidance.
    About HuMax-AXL-ADC
    HuMax-AXL-ADC is an ADC targeted to AXL, a signaling molecule expressed on many solid cancers and implicated in tumor biology. HuMax-AXL-ADC is in Phase I/II clinical development for seven different solid tumors: ovarian, cervical, endometrial, thyroid, non-small cell lung cancer (NSCLC), melanoma, and sarcoma. HuMax-AXL-ADC is fully owned by Genmab and the ADC technology used with HuMax-AXL-ADC was licensed from Seattle Genetics.

    Signed in September 2014, Genmab utilizes Seattle Genetics’ auristatin-based ADC technology with Genmab’s HuMax®-AXL, an antibody targeting AXL. Seattle Genetics received an initial upfront payment of USD 11 million and is entitled to receive more than USD 200 million in potential milestone payments and mid-to-high single digit royalties on worldwide net sales of any resulting products. In addition, prior to Genmab’s initiation of a Phase III study for any resulting products, Seattle Genetics has the right to exercise an option to increase the royalties to double digits in exchange for a reduction of the milestone payments owed by Genmab. Irrespective of any exercise of option, Genmab remains in full control of development and commercialization

    Goedemorgen mede-Genmabbers het eerste halfjaar van 18 zit erop en we kunnen niet anders concluderen dan dat het een teleurstellend eerste half jaar was . De teller staat op 7% in de min laten we hopen dat ze gas gaan geven in H2 want dit schiet niet op weer een jaar met negatief rendement . Kom op Dames & Heren jullie hebben een nieuw leuk (hopelijk zeer inspirerend) werkomgevinkje doe je best !
